ΜΕΤΑΛΛΑΞΟΓΕΝΕΣΗ ΚΑΙ ΕΝΕΡΓΟΠΟΙΗΣΗ ΤΟΥ RAS ΟΓΚΟΓΟΝΙΔΙΟΥ ΑΠΟ ΑΛΚΥΛΙΩΤΙΚΟΥΣ ΚΑΡΚΙΝΟΓΟΝΟΥΣ ΠΑΡΑΓΟΝΤΕΣ
Abstract
dc:description06-METHYLQUANINE, THE MAJOR PREMUTAGENIC AND PRECARCINOGENIC LESION INDUCED BY METHYLATING AGENTS, WAS INTRODUCED, BY MEANS OF SITE-DIRECTED MUTAGENESIS, INTOTHE CODONS 12 AND 13 OF THE HUMAN HA-RAS1 PROTO-ONCOGENE. THE 06MEG MUTAGENESIS WAS STUDIED IN E. COLI CELLS OF DIFFERENT REPAIR CAPACITIES: 1) O6MEG INDUCEDEXCLUSIVELY G-> A MUTATIONS, 2) THE MECHANISM OF THE METHYL- DIRECTED MISMATCHREPAIR PROTECTS, INDIRECTLY THE E. COLI CELL FROM THE O6MEG MUTAGENESIS INDIRECTLY, BECAUSE IT REPAIRS THE G:T DEMETHYLATION 3) IN ALKYLTRANSFERASE DEFICIENTE. COLI CELLS, THE O6MEG MUTAGENIC EFFICIENCY IS SIGNIFICANTLY INCREASED, THEREFORE THE ALKYLTRANSFERASE IS THE MAJOR REPAIR ENZYME OF THE LESION. HOWEVER, IN THE ABSENCE OF ALKYLTRANSFERASE AND MISMATCH REPAIR, THE MAIN EXCISION REPAIRWHICH IS PROVED TO HAVE A REMARKABLE EFFICIENCY TOWARDS O6MEG, SINCE THE MUTAGENIC EFFICIENCY NEVER REACHED HIGHER THAN 10-23%.
