Αντισώματα κατά Ro(SSA) και La(SSB) κυτταρικών αντιγόνων: Ανάπτυξη μεθόδου ανοσοαποτύπωσης (Western blot)

An immunoblotting method for the detection of antiRo and antiLa antibodies was developed, using two antigenic sources, thymus extract and human red blood cells. The reactivity of sera from patients with primary Sjogren’s syndrome towards the Ro and La polypeptides was examined and compared with the detection of precipitins by counterimmunoelectrophoresis (CIE). Sera with antiRo reactivity recognized two polypeptides with MW 50KD and 60KD and sera with antiLa reactivity one polypeptide with 40KD. Immunoblotting was more sensitive in the detection of antiRo (89%) and antiLa (90%) antibodies than CIE (76% and 73% respectively). AntiLa antibodies were frequently detected in sera of patients with antiRo precipitins antiLa antibodies were detected in sera of patients with antibodies to the 50KD Ro polypeptide rather than in sera of patients with antibodies to the 60KD Ro polypeptide indicating that La and 50KD Ro polypeptides are interrelated. Patients with antibodies to the 50KD Ro and La antigen exhibited a severe form of the disease.
