A HIGH PERFORMANCE LIQUID CHROMATOGRAPHIC (HPLC) METHOD FOR THE DETERMINATION OF PENICILLIN G, PENICILLIN V, OXACILLIN AND CLOXACILLIN IN COW'S MILK, HAS BEENDEVELOPED. PENICILLINS ARE EXTRACTED FROM MILK WITH DICHLOROMETHANE. CLEAN UP IS ACCOMPLISHED BY PARTITIONING BETWEEN BUFFERS AND ORGANIC SOLVENTS . PENICILLINS ARE FINALLY EXTRACTED INTO CHLOROFORM AS ION PAIRS WITH TETRABUTYLANNONIUM CATIONS. FOR HPLC, A REVERSED-PHASE COLUMN (NUCLEOSIL 5 C18) IS USED. THE MOBILE PHASE IS A MIXTURE OF 0,02M PHOSPHATE BUFFER AND ACETONITRILE (62:38,V/V), PH6, CONTAINING 5 MM TETRABUTYLAMMONIUM HYDROGEN SULFATE. THE ACCURACY OF THE METHOD IS 82,9+3,3% FOR PENICILLIN G, 89,3+5,1% FOR PENICILLIN V, 90,7+2% FOR OXACILLIN AND 90,6+4,6% FOR CLOXACILLIN, WHILE PRECISION DATA SHOW AN OVERALL RELATIVE STANDARD DEVIATION OF 4,61%, 6,56%, 3,77%, AND 5,83% FOR PENICILLIN G, PENICILLIN V, OXACILLIN AND CLOXACILLIN, RESPECTIVELY. THE LINEARITY OF THIS METHOD, WHICH PROVIDES QUANTITATION DOWN TO 4PPB FOR PENICILLIN G AND PENICILLIN V AND 3PPB FOR OXACILLIN AND CLOXACILLIN, IS EXCELLENT (R-2>0.998) IN THE RANGE OF 4-100PPB.
