AN ACIDIC, A NEUTRAL AND AN ALKALINE RIBONUCLEASE WERE ISOLATED FROM MOUSE LIVER CYTOPLASM, BY GEL FILTRATION, ION EXCHANGE AND AFFINITY CHROMATOGRAPHY. THE ACIDIC RIBONUCLEASE HYDROLYSES RNA OPTIMALLY AT PH 6.2, IN THE ABSENCE OF SALTS AND AT PH 5.5 IN SOLUTIONS WITH 150 MM OF MONOVALENT SALTS. IT IS NOT AFFECTED BY MG+2, ATP AND EDTA, BUT IT IS INHIBITED BY CA+2 AND ZN+2. IT IS ACID RESISTANCE AND HEAT LABILE. IT HYDROLYSES POLYPYRIMIDINORIBONUCLEOTIDES AND RNA IN NP-U AND GPAP-G BONDS. THE NEUTRAL RIBONUCLEASE HAS OPTIMUM PH 7.5 AND NEEDS 100-200 MM OF MONOVALENT SALTS. IT IS NOT AFFECTED BY ATP AND EDTA AND IS INHIBITED BY MG+2, CA+2 AND ZN+2. IT IS ACID AND HEAT RESISTANT. IT HYDROLYSES POLYPYRIMIDINORIBONUCLEOTIDES AND PYRIMIDINE-PURINE AND PYRIMIDINE-PYRIMIDINE BONDS IN RNA. THE ALKALINE RIBONUCLEASE HAS OPTIMUM PH 8.5, IN THE ABSENCE OF SALTS AND PH7 IN BUFFERS WITH 150 MM MONOVALENT SALTS. IT IS NOT AFFECTED BY ATP AND EDTA AND IS INHIBITED BY MG+2, CA+2 AND ZN+2. IT IS ACID AND HEAT RESISTANT. IT HYDROLYSES POLYPYRIMIDINORIBONUCLEOTIDES AND MAINLY POLY(C), RNA IS HYDROLYSED BY THIS ENZYME, IN THE SAME WAY AS BY THE NEUTRAL AND THE PANKREATIC RNASE. ALL THREE ENZYMES ARE ENDONUCLEASES AND THEY PRODUCE 3' PHOSPHOMONOESTERS. THEY DO NOTHYDROLYSE SS OR DS DNA. IN THE ABSENCE OF MONOVALENT SALT, THEY ARE ATTACHED STRONGLY ON THE POLYSOMES.
