ΣΥΜΒΟΛΗ ΣΤΗ ΜΕΛΕΤΗ ΚΑΙ ΤΟ ΧΑΡΑΚΤΗΡΙΣΜΟ ΤΗΣ ΕΞΩΚΥΤΤΑΡΙΑΣ ΟΥΣΙΑΣ (SLIME) ΒΑΚΤΗΡΙΑΙΜΙΚΟΥ ΒΛΕΝΝΩΔΟΥΣ ΚΑΙ ΘΥΓΑΤΡΙΚΟΥ ΜΗ ΒΛΕΝΝΩΔΟΥΣ ΣΤΕΛΕΧΟΥΣ PSEUDOMONAS AERUGINOSA
Abstract
dc:descriptionTHE FIRST CASE OF A HEAVILY MUCOID STRAIN (MS) OR PSEUDOMONAS AERUGINOSA BACTEREMIA IS REPORTED IN A HOSPITALIZED 78-YR-OLD WOMAN WITH DRUG-INDUCED HEPATITIS DUE TO ANTITUBERCULOUS THERAPY. A REVERTANT NON MUCOID VARIANT (RNMV) FROM THE PARENTAL MS WAS OBTAINED BY SERIAL PASSAGE ON TRYPTICASE SOY AGAR. THE EXTRACELLULAR MATERIAL (SLIME) FROM BOTH STRAINS WAS EXTRACTED BY METHODS AS FOR THE EXTRACTION OF GLYCOLIPOPROTEIN (GLP). GROSS CHEMICAL ANALYSES OF THE MS'S AND RNMV'S SLIME FOR URONIC ACIDS, CARBOHYDRATES, PROTEINS AND LIPIDS WERE 65%, 5.5%, 1.0%, 0.9% AND 16%, 48%, 11% AND 2% OF DRY WEIGHT, RESPECTIVELY. ANTIBODIES TO THE EXTRACELLULAR MATERIAL OF BOTH STRAINS WERE RAISED IN NEW ZEALAND WHITE RABBITS AND CROSS REACTIVITY IN HIGH TITERS AMONG THEM WAS SHOWN BY PASSIVE HEMAGGLUTINATION. ALSO, IT WAS SHOWN THAT THE EXTRACELLULAR MATERIAL OF THE RNMV EXERTED MITOGENIC ACTIVITY ON HUMAN PERIPHERAL LYMPHOCYTES WHILE THE MS'S EXTRACELLULAR MATERIAL WAS NOT MITOGENIC. THE MAJOR COMPONENT OF THE MS'S SLIME WAS ISOLATED BY ION EXCHANGE CHROMATOGRAPHY (IEC) AND GEL FILTRATION. IT WAS CHARACTERIZED, BY GELATINIZATION WITH CACL2, VISCOCITY MEASUREMENTS AND INFRARED SPECTROSCOPY, AS ALGINATE OF HIGH MOLECULAR WEIGHT, SIMILAR TO THAT PRODUCED BY MUCOID P.AERUGINOSA STRAINS FROM CYSTIC FIBROSIS. UNDER THE SAME CONDITIONS, IEC OF THE RNMV'S MATERIAL REVEALED TWO MAJOR PEAKS OF WHICH THE FIRST WAS COMPOSED OF NEUTRAL SUGARS (NS) AND PROTEIN IN ABOUT 5:1 RATIO, AND THE SECOND WAS CONSISTEDOF BOTH NS AND URONIC ACIDS. THIS URONIC ACID WAS SEPARATED FROM THE NS MOIETYBY PRECIPITATION WITH CACL2 AND WAS ALSO CHARACTERIZED AS ALGINATE BY INFRAREDSPECTROSCOPY. UREA -SDS- POLYACRYLAMIDE GEL ELECTROPHORESIS SHOWED THAT THE CELLS OF BOTH MUCOID AND NON MUCOID P.AERUGINOSA STRAINS CONTAIN SMOOTH LIPOPOLYSACCHARID THE RNMV'S SLIME DID NOT BEHAVE AS LIPOPOLYSACCHARIDE AS SHOWN BY UREA-SDS POLYACRYLAMIDE ELECTROPHORESIS AND BY LIMULUS AMOEBOCYTE LYSATE PYROTEST.
