THIS THESIS IS PART OF THE SCIENTIFIC PROGRAM OF OUR LABORATORY WHICH STUDIES THE TRANSLATIONAL CONTROL DURING THE DEVELOPMENT OF THE INSECT CERATITIS CAPITATA. IN THE FIRST PART OF THIS WORK THE METHOD OF PURIFICATION OF A NEW RIBONUCLEASE IS DEVELOPED. VARIOUS PROPERTIES ARE STUDIED, TOGETHER WITH HER ACTION IN IN VITRO SYSTEMS OF PROTEIN SYNTHESIS. AMONG THE CONCLUSIONS ARE THE FOLLOWING: A) THE RIBONUCLEASE THAT IS ISOLATED FROM THE LARVAE OF CERATITIS CAPITATA IS A THERMOSABILE MOLECULE WITH MOLECULAR WEIGHT OF 25000 DALTONS. B) THIS ENZYME DEGRADES SPECIFICALLY POLY(U) AND POLY(C) WHEREAS IT HAS NO EFFECT ON POLY(A), POLY(G) AND POLY(U,C). IT HYDROLYSES THE 18S AND 28S RIBOSOMAL RNA TO VARIOUS SPECIES OF RNAS WHICH ARE NOT HYDROLYSED ANY FURTHER. IN THE SECOND PART OF THIS WORK THE METHOD OF PARTIAL PURIFICATION OF A FACTOR WHICH INHIBITS THE ELONGATION OF NASCENT POLYPEPTIDE CHAINS OF POLYRIBOSOMES IS DEVELOPED ANDHS PROPERTIES TOGETHER WITH ITS ACTION IN IN VITRO SYSTEMS OF PROTEIN SYNTHESIS ARE STUDIED.
