ΜΕΛΕΤΗ ΤΗΣ ΕΠΙΔΡΑΣΗΣ ΤΟΥ PAF ΣΤΟ ΚΥΚΛΟΦΟΡΙΚΟ ΣΥΣΤΗΜΑ ΤΩΝ ΕΠΙΜΥΩΝ ΚΑΙ ΣΤΗ ΡΥΘΜΙΣΗ ΤΗΣ ΜΕΤΑΦΟΡΑΣ ΙΟΝΤΩΝ ΣΤΟ ΠΡΩΤΟΖΩΟ TETRAHYMENA PYRIFORMIS
Abstract
dc:descriptionTHE EFFECT OF THE SYNTHETIC PAF (AGEPC) ON THE CIRCULATORY SYSTEM OF RATS AND PARTICULARLY ON ECG AND THE EFFECT OF AGEPC ON THE REGULATION OF THE INTRACELLULAR LEVELS OF THE IONS CA2+, NA+ AND K+ WHICH PLAYS A MAJOR ROLE ON THE HEART FUNCTION WERE STUDIED. FOR THE BETTER EXPLANATION OF THE AGEPC ACTION TWO DRUGS (VERAPAMIL AND DIGOXIN) WERE USED AS POSSIBLE INHIBITORS AND WHICH ARE OFTEN USED IN CARDIAC DISEASES. IN THIS STUDY THE CELL OF THE PROTOZOO TETRAHYMENA PYRIFORMIS WAS USED. AFTER I.V. ADMINISTRATION OF AGEPC IN RATS, IT CAUSES HAEMOCONCENTRATION, ABRAPT FALL OF THE SYSTOLIC BLOOD PRESSURE AND CARDIAC ISCHAMIA WHICH RESULT IN INFARCT WITH LETHAL DOSES OF AGEPC. THIS EFFECT OF AGEPC SEEMS TO BE THE RESULT OF A DIRECT VASCOCONSTRICTING ACTION ON CORONARY VESSELS. AGEPC ALSO CAUSES AN ABRAPT INCREASE OF INTRACELLULAR FREE CA2+ AND A SLOW INCREASE OF INTRACELLULAR NA+ BUT IT HAS NOT ANY EFFECT ON INTRACELLULAR LEVELS OF K+ AND ON CELL'S VOLUME. THIS EFFECT OF AGEPC IS COMPLETELY INHIBITED BY VERAPAMIL AND PARTIALLY BY DIGOXIN. THE NON ACETYLATED DERIVATIVE OF AGEPC LYSO-GEPC HAS NOT ANY ONE FROM THE ABOVE MENTIONED EFFECTS OF AGEPC.
