AN INVESTIGATION OP POSSIBLE IN VITRO METHODS FOR STUDYING EYE IRRITANCY POTENTIAL

Two in vitro cytotoxicity procedures, namely the measurement of cell membrane integrity using fluorescein diacetate and ethidium bromide, and the quantitation of the release and activity of a cell membrane-bound enzyme, alkaline phospha1ase,were used to assess the qytotoxicity of a range of cationic, anionic and nonionic detergents. The in vitro results were compared with the in vivo irritancy of these compounds in the rabbit eye and the significance of any correlations are discussed. In addition, measurements were made of detergent penetration through the isolated. rabbit cornea and the rates of change of in vitro corneal electrical impedance in the presence of detergents. These findings are also compared with the in vivo results. The composite effects of intrinsic cellular damaging potential, penetration effects on proteins and adsorption of detergents are discussed as they relate to a model of in vivo ocular irritancy.
