The Effect of Primary Hepatocyte Expressing HBx on the Activation and Apoptosis of Antigen-specific CD8+ T Cell
Abstract
dc:descriptionCD8^(+) T cells contribute to the control of Hepatitis B virus (HBV) infection by killing of infected cells or inhibiting viral replication inside cells. HBx, antigen of hepatitis B virus, plays an important role in the immunopathogenesis of HBV infection. Since HBx is able to upregulate cellular expression level, it is thought that HBx may modulate CD8^(+) T cell immune response. To test this hypothesis, proliferation, cytokine production and apoptosis of CD8^(+) T cells co-cultured with hepatocytes expressing HBx were analyzed. First, recombinant baculovirus containing mammalian gene expression cassette for X gene was generated. Then, female MataHari naive CD8^(+) T cells were cultured with C57BL/6 mouse primary hepatocytes infected with baculovirus expressing HBx. At the end of co-culture, cell proliferation, cytokine production and apoptosis of CD8^(+) T cells were analyzed. As the results, the cell proliferation of naive CD8^(+) T cells co-cultured with primary hepatocytes expressing HBx was equivalent with naive CD8^(+) T cells co-cultured with normal hepatocytes. CD8^(+) T cells co-cultured with primary hepatocytes expressing HBx was shown a increase in the IFN- r production but this result made no sens. But apoptosis was increased in CD8^(+) T cells co-cultured with hepatocytes expressing HBx. In conclusion, this results suggest that primary hepatocytes infected HBV can modulate cytotoxic T lyphocytes immune response. This study will improve understanding of the mechanism how to chronic hepatitis and hepatocellular carcinoma develop and might provide a new sight of the treatment of HBV infection.
