Suppressive effect of a nucleic acid-hydrolyzing recombinant antibody (3D8) on viral multiplication

3D8 single chain variable fragment (scFv) is a recombinant anti-DNA antibody from an autoimmune-prone MRL-lpr/lpr mouse with lupus diseases. 3D8 scFv can hydrolyze ss-DNA, ds-DNA, ss-RNA, and ds-RNA without sequence specificity. On the base of 3D8 scFv property, we carried out quantitative and qualitative analyses of antiviral activity of 3D8 scFv against vesicular stomatitis virus (VSV) that belongs to RNA viruses in HeLa cells. When the purified 3D8 scFv was directly imported into HeLa cells by an electroporation and the cells were infected with VSV (MOI=10), significant inhibition against VSV multiplication and VSV-induced cytopathic effect (apoptosis) was observed. When HeLa cells that stably express 3D8 scFv protein were infected with VSV (MOI=0.1), 3D8 scFv-expressing cells showed 40% reduction in the viral G protein expression level with flow cytometry and 1 x 10³ reduction of viral particle yield with plaque forming assay, compared to the control HeLa cells. We found that intracellularily expressed 3D8 scFv retains nucleic acid-hydrolyzing activity and type I IFN induction was not more sensitized by VSV infection in the HeLa cells expressing 3D8 scFv, when compared to the control cells. It suggests that antiviral effect due to the nucleic acid-hydrolyzing activity of 3D8 scFv. In conclusion, we confirmed that nucleic acid-hydrolyzing 3D8scFv exhibits antiviral activity to VSV in the HeLa cells.
