Identification and Analysis of Peanut and Soybean Cross Reacting Allergens in Korean Children Sensitive to Peanut

BACKGROUND: Peanut and soybean are among the most prevalent allergens of food hypersensitivity. They both belong to legume family and share high similarity in many parts of their seed storage proteins. the clinical manifestations of allergy to peanut and soybean are generally quiet distinct. And also, there are many cases of allergic patients with high level of soybean-IgE without clinical soybean allergy, especially in patients with high level of peanut-IgE. OBJECTIVE: The aim of this study was to evaluate the IgE binding proteins in both species, with a goal of identifying homologous epitopes of peanut and soybean protein that can potentially elicit serious allergic reactions. METHODS: Sera were obtained from 17 patients with high levels of peanuts and soybean specific IgE antibodies by ImmunoCAP(Pharmacia Diagnostics). Unabsorbed sera and sera adsorbed to remove cross-reacting antibodies were assayed for specific IgE binding to peanut or soybean immunoblots. SDS-PAGE and peanut-IgE immunoblotting with soybean-inhibited peanut allergy patient sera and non inhibited patient sera were performed. The spot membrane containing the peptides was analysed by Procise 492 cLC Protein sequencer (Applied Biosystem, Foster City, CA). Peptide synthesis of predicted Peanut-Soy common IgE binding epitopes were performed and then predicted epitope IgE ELISA to peanut hypersensitive patient's sera were performed. RESULTS: IgE inhibition ELISA test using peanut sensitized sera showed significant inhibitions by soybean protein at 800ug concentration. Using the ELISA inhibition test, there are some degree(25.15%) of cross reactivity between epitope #1 and soybean antigens was noted. The results of IgE inhibition immunoblot with peanut hypersensitive patients' sera inhibited to soybean glycinin disappeared several protein bands, especially two bands was prominent(38 kDa, 23 kDa). A search for sequence homology to the protein spots revealed that significant homology to peanut Ara h 3. CONCLUSION: This study has identified a common IgE-binding epitope site present with the peanut Ara h 3(38 kDa, 23 kDa) and soybean glycinin proteins. This site could contribute to the cross-reactions observed in sera between peanut- and soybean-sensitive individuals.
