Effects of loigomycin, F0/F1 ATP-synthase inhibitor on mitochondrial morphology and dynamics

dc:descriptionMitochondrial fission has recently been well-recognized as a factor involved in apoptosis although its role and the underlying mechanism remains unclear. Recently, we monitored the effects of mitochondrial respiratory inhibitors [rotenone, TTFA (2-thenoyltrifluoro-acetone), antimycin A, and oligomycin] to investigate the complex-specific retrograde signal pathways. The inhibitors showed different effects on mitochondrial dynamics (distribution and morphology), but all accompanied apoptosis. Among the respiratory inhibitors, oligomycin induced rapid mitochondrial fragmentation within 5 minutes and the irregularly fragmented forms of mitochondria maintained with progressive decrease of the mass until the cells die. We found that the intracellular ATP level reduced by oligomycin did not affect on the mitochondrial fission, but high ROS level produced in early stage modulate mitochondrial fission machinery, which is evidenced by blocking of the fission with antioxidants. Finally, the early mitochondrial fission induced by oligomycin was independent of cytochrome c release from the mitochondria. Our data suggest that oligomycin-induced mitochondrial fission dose not directly related to opening of mitochondrial apoptotic signaling and ROS is an important mediator in the fission process.
