Untersuchung der nichtkodierenden Region (NCR) von HPV 6 an Paraffinschnitten von Condylomata acuminata

The paper submitted examins whether structural mutations within the non-coding region of HPV 6 can be found in benign Condylomata acuminata, and how frequently these mutations can be found in these benign lesions. Malignant Condylomata acuminata und Buschke-Loewenstein Tumors with dublication mutations in the non coding region of HPV 6 were described in the literature. This led to the suspicion that mutations increase the malignant potential of HPV 6. 96 formalin embedded Condylomata acuminata of 54 patients were examined. We used the polymerase chain reaction combined with restriction enzymes analysis to detect mutations within the non-coding region. Embedding tissue samples in paraffin leads to damage of the DNA, therefore we used gene clean® to clean the DNA of fragments and a more sensitive PCR (hemi-nested-PCR). Using this combination of techniques we were able to detect viral DNA in 28 of 96 Condyloma acuminata, amplify it and analyse it. In one of these clinically benign samples we found a restriction enzyme pattern that could be explained by a tandem-duplication of about 200bp length in the 3' region of the non-coding region of HPV 6. Later on we were able to show that this sample indeed included an exact tandem duplication of a part of the non-coding region. Mutations within the non-coding region of HPV 6 seem thus to be rare events in Codylomata acuminata but are not restricted to Buschke- Loewenstein tumors or HPV 6 induced squamous cell carcinomas. The biological significance, however, cannot be evaluated by epidemiological data alone.
