QUB-1957: a bioactive peptide from the frog skin secretion of Hylarana latouchii
Abstract
dc:description.abstractThe skin secretions of amphibians are significant for amphibians’ daily protection, especially the antimicrobial peptides. In this study, a cDNA library from lyophilised skin secretion of Hylarana latouchii was constructed, and molecular cloning of biosynthetic precursor-encoding cDNAs of QUB-1957 was performed. Subsequently, QUB-1957 was synthesised by Solid Phase Peptide Synthesizer. In the antimicrobial assays, the MIC of QUB-1957 against Gram-positive bacteria Staphylococcus aureus, Gram-negative bacteria Escherichia coli and yeast Candida albicans are 8 μM, 16 μM and 16 μM, respectively. Unfortunately, QUB-1957 also exhibits a certain degree of haemolytic rate, the HC50 is 47.82 μM. Besides, QUB-1957 exhibits no significant effect against PC3 (human prostate cancer cell line), NCI-H157 (human non-small cell lung cancer cell line), MCF-7 (human breast cancer cell line) and U251MG (human neuronal glioblastoma cell line). These characteristics of QUB-1957 indicate that QUB-1957 is a promising antimicrobial agent with a broad spectrum that worth further study. Hopefully, the structure QUB-1957 could be researched for novel antimicrobial agent development.
