Abstract
dc:descriptionWuhan Mosquito Virus 6 (WuMV-6) is a member of the Orthomyxoviridae family, first discovered in China in 2013. Its rapid migration and high genetic variability raise significant questions about its epidemiology and potential threat to vertebrates. The rapid evolution of the virus's surface protein gp64 and the limited flight range of mosquitoes suggest a possible role of birds as hosts for the virus. This study aimed to detect WuMV-6 in Culex pipiens mosquito samples collected in Lithuania. Specific primers were developed and validated for detecting the hypothetical segment 1 and the nucleoprotein segment of the WuMV-6 genome. Using advanced molecular methods, such as polymerase chain reaction (PCR) and sequencing, specific segments of the WuMV-6 genome were identified. The study confirmed that WuMV-6 circulates in local C. pipiens mosquito populations in Lithuania, with nine independent virus detection cases found among sixty-one examined samples. Additionally, phylogenetic analysis revealed that the detected WuMV-6 segments are not endogenous viral elements (EVEs), indicating that the detected viral RNA originated from active infections rather than genetic remnants. Phylogenetic analysis also revealed that WuMV-6 found in Lithuania is genetically closest to variants found in Sweden and has more distant relationships with variants spread in other regions of the world. These results expand our understanding of WuMV-6 evolution and its global distribution. However, further research is needed to determine the transmission routes of WuMV-6 in mosquito populations and what its host range is.
