Based on the changes in gene expression over time and the effect of chloroquine, the limiting step of hybrid vector cell transduction appears to be endolysosomal escape of the complex. The polymer component fails to mediate escape as efficiently as the MLV envelope protein, likely resulting in the degradation of internalized hybrid vector complexes.
