Tyrosine kinase inhibitors block signals involved in cell growth and differentiation and are often used in the treatment of cancer. Previous phase I research demonstrated that a high-dose intermittent schedule of sunitinib is safe and feasible, while achieving higher drug concentrations in the tumor. This regimen was studied in two clinical trials in patients with glioblastoma and metastatic colorectal cancer. However, treatment with high-dose sunitinib did not result in a significant improvement in progression-free survival or overall survival compared to standard treatment. A biomarker study performed with samples from colorectal cancer patients highlighted the potential of circulating tumour DNA (ctDNA). These are small fragments of genetic material derived from the tumour which circulates in the bloodstream. Measuring ctDNA shows promise for predicting treatment response and providing more insight into patient prognosis.","Radboud University, 15 april 2025","Promotores : Verheul, H.M.W., Medema, J.P.
