This thesis examined neurochemical status and potential therapeutic roles of Histaminergic H3 and Cannabinoid CB1 receptors in neurodegenerative diseases. Based on neurochemical investigations in postmortem human brain samples, the status of H3 receptors remain unchanged under vascular dementia or mixed dementia pathology, and may serve as a therapeutic target for cognitive deficits. Measurements of CB1 receptors in postmortem brain samples of Lewy body dementia patients indicate the possible relationship between CB1 loss and amyloid plaque burdens as well as synaptic deficits. Furthermore, the effect of CB1 inverse agonists suppressing the non-amyloidogenic APP cleavage mediated by muscarinic agonist in SH-SY5Y neuroblastoma cell line suggest the loss of protection against A ß pathology when CB1 inverse agonists are co-administrated with cholinergic neurotransmission enhancements.
