SYNTHESIS OF NOVEL BODIPY LIBRARIES AND THEIR APPLICATION IN BIOIMAGING

The need for recognizing and sensing environmentally and biologically important species has leaded large efforts towards sensor development. We first applied a combinatorial approach to construct diversity-oriented fluorescent libraries as chemical tool box based on BODIPY fluorophore. These novel BODIPY libraries were then carried out in both unbiased in vitro screening and cell-based screening systems. Novel sensors towards fructose, glutathione, date-rape drug-GBL, as well as neural stem cells, were identified from each screening processes, and evaluated to show good application in detection or quantification of specific target. This diversity-oriented approach was also used to study the structure and cellular behavior relationship of BODIPY dyes in order to ¿tame¿ them. In later development of a protein tag system based BODIPY diacrylate structure, this information was successfully used to solve the background issue of the dye. These work discovered useful biological and environmental sensors, and provided information on protein tag development.
