PAMAM dendron, a hyperbranch of the PAMAM dendrimer, was divergently synthesized on solid support. Prior to the propagation through (1) Michael addition of acryl ester to an amino reactive site of the organic phase grafted onto the silica gel and (2) amidation of the resulting ester moieties with alkylene diamine, the silica particles were treated with 3-aminopropyltriethoxysilane that would serve as a coupling agent. Afterwards, the cleavage of the solid-phase synthesized full generation PAMAM dendrons had been done to perform structural analysis including determination of synthesis defects. The cleavage is considered a very important part of the synthesis because of the numerous applications of the macromolecule in the liquid phase. To date, the cleavage of PAMAM dendron from the silica gel has not been accomplished previously.
