DESIGN AND CONSTRUCTION OF BIOSENSING PLATFORMS FOR THE DETECTION OF BIOMARKERS
Abstract
dc:description.abstractThis study aims on the development of biosensors for the detection of two biomarkers, glucose and DNA methyltransferase (MTase), which have been recognized as the target for diabetes mellitus and aberrant DNA methylation related diseases including cancer, respectively. On the one hand, three types of electrochemical glucose biosensors have been constructed by using redox polymers and lamellar-ridge architectured gold. On the other hand, three different signal generation strategies have been employed for DNA MTase activity analysis: the first one uses a synthetic threading intercalator, N,N?-bis(3-propylimidazole)-1,4,5,8-naphthalene diimide functionalized with Os(bpy)2Cl+ moieties, which catalyzes the electro-oxidation of ascorbic acid; the second one relays on a MoS2 nanosheet-mediated fluorescence quenching-restoration strategy; and the last one adopts a personal glucose meter as the signal readout through the conjugation of invertase with the substrate DNA of MTase. All of these established biosensors show good analytical performances, and may potentially be useful tools for clinical applications.
