This system is based on a confocal-multiphoton-laserscanning microscope and comprises an optical laser-tweezers module, total-internal-reflectance microscopy (TIRFM), an additional stationary multi-photon coupling and position detectors. The CMLTT system is designed so that each module can be used with others in parallel. The laser beam forming the optical tweezers can be modulated both in space and time using acoustooptical and electrooptical modulators. The TIRFM module is built to accustom two excitation light sources and the power coupling of the stationary multi photon module can be varied. For laser tweezers applications, three position detectors are available for all microscopic modes. This modular CMLTT microscopic apparatus has been tested on myofibrillar preparations and single molecule assays. For the first time, it was possible to carry out high resolution second harmonic measurements of the three dimensional sarcomeric structure of muscle cells. Additionally, the multi photon photo release of DMNPE-4, a calcium cage molecule, has been realized.
