The objective of this research was to evaluate host responses induced by MCWF in canine peripheral blood mononuclear cells (PBMCs), using enzyme-linked immunosorbent assays (ELISA) and quantitative reverse-transcription polymerase chain reaction (qRT-PCR) for quantifying cytokine protein and mRNA transcripts, respectively. PBMCs were isolated from eight healthy dogs and cells were cultured with MCWF. A significant increase of interleukin (IL)-10 mRNA was detected at all time points for all concentrations of MCWF (p<0.05). mRNA relative abundance reflected this finding. A post-stimulation elevation of interferon (IFN)-γ mRNA was noted at 24-hours with all concentrations of MCWF (p<0.01) and tumor necrosis factor (TNF)-α mRNA was increased for 0.5ng/dL MCWF only at 72-hours (p<0.05) compared to the negative control.
