Abstract
dc:descriptionThe effect of VPA on the process of puberty, on conditioned avoidance behaviour learning and on brain neurotransmitters of the rats was studied, after chronic and acute administration of the drug and after stress. The brain neurotransmitters NA, DA, DOPAC, (DOPAC/DA), 5-HT, 5HIAA, (5HIAA/5-HT) were measured with High Liquid Pressure Chromatography (HLPC) on the Frontal Cortex (FC), Ypothalamus (Y) and Striatum (S, on which were measured HVA and HVA/DA as well). Measurements of neurotransmitters on these three brain regions were done after chronic (per os for 15-27 days, at dose 100-200 mgr/ Kgr), or acute (intraperitonal 200 mgr/Kgr) treatment with VPA on prepubertal male and female rats. 48 hours before decapitation and measurement of brain amines, some groups of animals were evoked on ether stress 15 min or test conditioned avoidance behaviour learning 30 min. In addition, the testosterone serum levels and the weight of organs of prepubertal male rats were studied, after acute VPA administration and stress. Strict statistical control was done that included checking the existence of normal division, t-test or Wilcoxon's test where the criteria of t-test had no effect. After the statistical evaluation of the data, it has been shown: 1. No remarkable difference was observed in any of the measures of puberty's process. 2. VPA caused decrease of concentration of DA on Y (P<0.05) ) on prepubertal rats, after chronic administration. However after a simple dose no remarkable differences of DA concentrations between VPA and control animals were observed. Increase of 5HIAA on FC (P<0.01) was shown after stress (48 hours before) and double ip dose of VPA (the first 30 min before stress and the second 30 min before decapitation) on prepubertal male rats, in comparison with animals that were evoked on stress and a simple dose of NaCL 0.9%. 3. After chronic administration of VPA and stress (48 hours before the death), decrease of NA concentration on FC and on Y and increase of 5-HT turnover was observed, as well as augmentation of 5HIAA on Y and on S, in comparison with control animals that received the drug chronic in same dose but they were not evoked by stress. 4. Stress (48 hours before the death of the animals), influenced the reduction of NA on FC and on S as well as the reduction of DA turnover on S; this happened to both the control and VPA rats (that received a double ip dose). Decrease of DOPAC on S was observed only on VPA rats; reduction of DOPAC and of ratio DAPOC/DA on Y were more remarkable on VPA animals (P<0.05). 5. The combination of double ip treatment of VPA and stress (48 hours before), in comparison with a simple administration of VPA without stress, caused reduction of NA concentration on FC and on S, augmentation of concentration of 5-HT on FC and of 5HIAA on FC, Y and S, as well as increase of 5-HT turnover on FC and on Y. No differences on serotoninergic system were observed after the comparison between control rats that were evoked by stress and control rats that were not evoked. 6. The test of ability of conditioned avoidance behaviour learning did not show any differentiation between the compared groups (VPA and control). Note that percentage avoidance was very low to both groups, so it is insecure to draw any conclusion. These data suggest that VPA does not effect the process of puberty, but it decreases the DA concentration on Y after chronic administration. Furthermore, it appears that stress influences remarkable the effect of VPA on serotonin turnover, in particular on Frontal Cortex. These actions of VPA on serotoninergic and dopaminergic system may be instrumental in the mechanisms of the anticonvulsant action of the drug.
