Monocytes, mainly lymphocytes, were isolated by the Ficoll-Hypaque system and cultivaled in RPMI-1640. The immune response of the lymphocytes was studied by adding in the culture medium hydatid fluid and measuring the [³H] thymidine incorporation into the DNA of the cells. The viability of scolices and lymphocytes was determined by the Trypan Blue exclusion technique. All experiments were carried out in triplicate. The findings of the study demonstrate: 1. Lymphocytes from patients with echinococcosis are stimulated when cultured in the presence of hydatid fluid. Maximum stimulation occurred on the sixth day of culture, at a hydatid fluid protein concentration of 15 μg per ml of culture medium. The stimulation Index (SI) of the lymphocytes obtained from 10 subjects with active or cured echinococcosis was 4.3 ± 1.6 (mean ± SD), with a range from 7.0 to 2.1. Lymphocytes from normal control subjects demonstrated a SI of 1.1 ± 0.3, with a range from 0.5 to 1.5. 2. The immune memory of the sensitized lymphocytes is maintained at least up to 20 years after removal of the hydatid cyst, even if the Casoni's skin test and serology have become negative. 3. The viability of E. granulosus scolices cultivated in the presence of sensitized lymphocytes was significantly lower than that of the same scolices cultivated in the presence of nonsensitized lymphocytes. On the 5th day of culture only 5% of the scolices cocultivated with sensitized lymphocytes were alive, whereas on the same day of culture 57%, of the same scolices cocultlvated under similar conditions with nonsensitized lymphocytes were alive. 4. Sensitized lymphocytes were killed by E. granulosus scolices to a much greater extend than nonsensitized lymphocytes. 5. The reduced viability of both sensitized lymphocytes and scolices, when cocultivated, was not the result of a nutrient depletion, but was caused by the lymphocyte immune response. 6. Η μεγαλύτερη λεμφοκυτταρική διέγερση παρατηρήθηκε την έκτη μέρα, με συγκέντρωση πρωτεΐνης υδατίδος κύστεως 15 mg/ml καλλιεργητικού υλικού. Ο δείκτης διεγερσιμότητας (ΔΔ) των λεμφοκυττάρων δέκα ατόμων με υδατίδα κύστη ή ιστορικό εχινοκοκκιάσεως ήταν 4,3 ± 1,6 (μέση τιμή ± ΣΑ), με διακύμανση από 7 έως 2,1. Στις καλλιέργειες μαρτύρων, ο Δ.Δ. ήταν 1,1 ± 0,3 (Μέση τιμή ΣΑ) με διακύμανση από 0,5 έως 1,5. Τα αποτελέσματα αποδεικνύουν ότι λεμφοκύτταρα ανοσοποιημένων ατόμων διατηρούν την αντιχονική μνήμη και εμφανίζουν βλαστική μεταμόρφωση in vitro παρουσία αντιγόνου. Η συγκαλλιέργεια σκωληκοκεφαλών Ε. Granulosus και ευαισθητοποιημένων λεμφοκυττάρων έδειξε ότι η ζωτικότητα τόσο των σκωληκοκεφαλών όσο και των ευαισθητοποιημένων λεμφοκυττάρων ήταν σημαντικά μικρότερη από εκείνη συγκαλλιεργειών σκωληκοκεφαλών με μη ευαισθητοποιημένα λεμφοκύτταρα. Σε συγκαλλιέργειες σκωληκοκεφαλών με ευαισθητοποιημένα λεμφοκύτταρα, το ποσοστό των ζωντανών σκωληκοκεφαλών την 5η ημέρα ήταν 5%, ενώ σε συγκαλλιέρχειες με μη ευαισθητοποιημένα λεμφοκύτταρα ήταν 57%. Ανάλογη μεγαλύτερη καταστροφή των λεμφοκυττάρων παρατηρήθηκε σε μικτές καλλιέργειες λεμφοκυττάρων και σκωληκοκεφαλών, όταν τα λεμφοκύτταρα προέρχονταν από άτομα, τα οποία είχαν νοσήσει ή νοσούσαν από εχινόκοκκο.
