Η ΕΠΙΔΡΑΣΗ ΤΗΣ ΧΡΟΝΙΑΣ ΧΟΡΗΓΗΣΗΣ ΔΙΦΑΙΝΥΛΟΔΑΝΤΟΙΝΗΣ ΣΤΗΝ ΠΩΡΩΣΗ ΠΡΟΚΛΗΤΩΝ ΚΑΤΑΓΜΑΤΩΝ ΤΗΣ ΚΑΤΩ ΓΝΑΘΟΥ ΕΠΙΜΥΩΝ
Abstract
dc:descriptionTHE PURPOSE OF THIS STUDY WAS TO INVESTIGATE WHETHER THE CHRONIC ADMINISTRATIONOF DIPHENYLHYDANTOIN (PHENYTOIN-NATRIUM) HAS ANY INFLUENCE ON THE PROCESS OF HEALING OF MANDIBULAR FRACTURES IN YOUNG RATS. FOR THIS PURPOSE 45 FEMALE WISTARRATS WERE USED SEPARATED IN THREE GROUPS OF 15 RATS EACH. TWO OF THESE GROUPS CONSISTED FROM THE EXPERIMENTAL ANIMALS WHILE THE THIRD ONE INCLUDED THE CONTROL ANIMALS. THE CHRONIC ADMINISTRATION OF DIPHYNYLHYDANTOIN WAS DONE BY I.P. INJECTION IN DOSES OF 10 AND 100 MG PER KG. OF BODY WEIGHT. THE ADMINISTRATION OF THE DRUG WAS STARTED 20 DAYS BEFORE THE SURGICAL PROCEDURE OF FRACTURE. THEN EACH GROUP WAS DIVIDED INTO THREE SUBGROUPS WITH 5 RATS EACH AND THE ADMINISTRATION OF THE DRUG WAS CONTINUED IN THE ABOVE MENTIONED DOSES PER GROUP OVER A PERIOD OF 7, 14 AND 21 POST- FRACTURE DAYS IN EACH SUBGROUP. ON THESE CORRESPONDING DAYS BLOOD SAMPLES WERE OBTAINED BY CATHETERIZING THE LEFT COMMON CAROTID ARTERY OF THE RATS, AFTER AN INTERVAL OF 12 HOURS FROM THE LAST ADMINISTRATION OF THE DRUG. FOLLOWING THIS EACH RAT WAS SACRIFICED BY INJECTING AN I.P. OVERDOSE OF PENTOBARBITAL SODIUM. THE MANDIBLES OF THE RATS WERE PREPARED FOR PARAFIN SECTIONS. DURING THIS EXPERIMENT THE 15 CONTROL ANIMALS OF THE THIRD GROUP WERE RECEIVING I.P. NORMAL SALINE TO THE SAME SCHEDULE. VALUES OF CALCIUM, PHOSPHORUSAND ALKALINE PHOSPHATASE WERE DETERMINED BY PHOTOMETRIC METHODS IN THE SERUM OF EVERY RAT. DIPHENYLHYDANTOIN WAS ALSO DETERMINED BY PHOTOMETRIC METHOD IN THESERUM OF THE RATS WHO WERE RECEIVING THE DRUG. IN THIS STUDY FRACTURE HEALING WAS ASSESED BY HISTOLOGICAL EXAMINATION AT 7, 14 AND 21 DAYS FOLLOWING THE FRACTURE. AN ATTEMPT WAS MADE TO CORRELATE THE FINDINGS OF THE HISTOLOGICAL EXAMINATION TO THE ADMINISTERED DOSES OF THE DRUG, TO THE DURATION OF THE DRUG'S ADMINISTRATION IN EACH GROUP AND TO THE VALUES OF CALCIUM, PHOSPHORUS AND ALKALINE PHOSPHATASE IN SERUM. COMPARING TO CONTROL ANIMALS, THE RESULTS OF THE PRESENT STUDY INDICATE THAT CHRONIC ADMINISTRATION OF DIPHENYLHYDANTOIN IN DAILY I.P. INJECTION OF 10 AND 100 MG PER KG. OF BODY WEIGHT IN YOUNG RATS, IN SPITE OF CAUSING SIGNIFICANT DECREASE OF CALCIUM AND PHOSPHORUS AND SIGNIFICANT INCREASE OFALKALINE PHOSPHATASE IN SERUM, DOES NOT ACCELARATE OR DELAY THE PROCESS OF HEALING AND DOES NOT CAUSE ANY QUALITATIVE ALTERATION ON THE HEALING, AS THIS WAS EXAMINED AT 7, 14 AND 21 POST- FRACTURE DAYS. SO IT COULD BE CONCLUDED THAT THEABOVE MENTIONED BIOCHEMICAL DIFFERENCES CAUSED BY DIPHENYLHYDANTOIN IN EXPERIMENTAL RATS HAVE NOT BIOLOGICAL SIGNIFICANCE ON HEALING OF EXPERIMENTAL MANDIBULAR FRACTURES IN THESE ANIMALS.
