Furthermore L-glutamate binding have been solubilized from the same membrane preparations using different detergents, as CHAPS, Zwittergent 3-12 and Digitonin. At last, a comparative study of L-[³H] glutamate binding sites was done between normal and OPCA (olivocerebellar atrophy) human cerebellum. The results of the above studies are as follows: 1. One binding component was found in both CI⁻ dependent and -independent conditions for L-glutamic acid with Kd values in the order of 150Χ10⁻⁹Μ. Hill plots of these data were linear with Hill coeficient equal to 1.0. Chloride ions provoked a significant increase of L-glutamate binding, with ratio of Cl⁻-dependent/-independent binding sites about 4/1. L-glutamate, L-aspartate, quisqualate, L-homocysteic and ibotenic acid were potent inhibitors of L-[³H] glutamate binding, whereas D,L-APB, kainic acid and NMDA were weak inhibitors. L-[³H] glutamate binding showed a significant degree of stereoselectivity toward L- and D-isomers of glutamate and aspartate. With the exception of quisqualate, all amino acid analogue used did not show significant differences in their affinities of inhibition between Tris-HCl and Tris-acetate buffer. Under Cl⁻ dependent conditions the inhibition curve of quisqualate was biphasic (IC50=6.30nM and 15,85μΜ), whereas under Cl⁻-independent conditions was monophasic with IC50 value very similar to the low affinity site observed in Tris-HCl buffer. The ontogeny of L-glutamic acid binding sites in human cerebellum showed that no significant changes of F' values were observed with age, whereas the highest Bmax value was observed at the age of 1 year. Autoradiography revealed L-glutamate binding sites in both granule cell and molecular layers of human cerebellum. [³H]-QNB binding expressed in fmoles/mg protein, did not show significant differences between normal and OPCA cerebella.","Στη παρούσα εργασία μελετήθηκαν οι ιδιότητες δέσμευσης, η επίδραση των ιόντων Cl⁻ στη δέσμευση, οι φαρμακολογικές ιδιότητες, η οντογένεση και η τοπογραφική κατανομή των θέσεων δέσμευσης του L-γλουταμικού οξέος σε μεμβρανικό κλάσμα παρεγκεφαλίδας ανθρώπου. Επίσης έγιναν πειράματα διαλυτοποίησης των θέσεων δέσμευσης του L-γλουταμικού οξέος με τη χρήση διαφόρων απορρυπαντικών, όπως το CHAPS, το Zwittergent 3-12 και η διγιτονίνη. Τέλος έγινε προσπάθεια συγκριτικής μελέτης των ιδιοτήτων της δέσμευσης του L-[³Η]γλουταμικού οξέος σε παρεγκεφαλίδα φυσιολογικών ατόμων και ατόμων που πέθαναν από ελαιογεφυρό-παρεγκεφαλιδική ατροφία. Τα αποτελέσματα των παραπάνω μελετών έχουν ως εξής: 1. Διαπιστώθηκε μια θέση δέσμευσης για το L-γλουταμικό οξύ, παρουσία ή απουσία ιόντων Cl⁻, με τιμές Κd της τάξης των 150 Χ 10⁻⁹. Η ανάλυση κατά Hill σε όλες τις περιπτώσεις έδωσε ευθεία γραμμή και η σταθερά Hill ήταν περίπου 1. Τα ιόντα Cl⁻ προκάλεσαν σημαντική αύξηση της δέσμευσης του L-γλουταμικού οξέος με αναλογία Cl⁻ εξαρτωμένων/ Cl⁻ ανεξάρτητων θέσεων δέσμευσης περίπου 4/1. Το L-γλουταμικό, L-ασπαρτικό, κισκαλικό, L-ομοκυστεικό και ιμποτενικό οξύ απεδείχθηκαν ισχυροί αναστολείς της ειδικής δέσμευσης του L-[³H] γλουταμικού οξέος στις μεμβρανικές θέσεις δέσμευσης του. Αντίθετα το D,L-APB, το καινικό οξύ και το NMDA ήσαν ασθενείς αναστολείς. Η δέσμευση του L-[³H] γλουταμικού οξέος έδειξε μια σημαντικού βαθμού στερεοεξειδίκευση μεταξύ των L- και D- ισομερών του γλουταμικού και ασπαρτικού. Με εξαίρεση το κισκαλικό, όλα τα ανάλογα αμινοξέα που χρησιμοποιήθηκαν δεν παρουσίασαν σημαντικές διαφορές της ικανότητας αναστολής παρουσία ή απουσία ιόντων Cl⁻. Παρουσία ιόντων Cl⁻ η καμπύλη αναστολής του κισκαλικού ήταν διφασική (IC50=6,30nM και 15,85μΜ), ενώ απουσία ιόντων Cl⁻ η καμπύλη αναστολής ήταν μονοφασική με τιμή IC50 παρόμοια της θέσης χαμηλής συγγένειας που παρατηρήθηκε παρουσία ιόντων Cl⁻. Η οντογενετική μελέτη της δέσμευσης του L-γλουταμικού οξέος στην ανθρώπινη παρεγκεφαλίδα έδειξε ότι δεν παρατηρούνται σημαντικές μεταβολές των τιμών της Kd με την ηλικία, ενώ η μεγαλύτερη τιμή Bmax παρατηρήθηκε τον 1ο χρόνο μετά τη γέννηση. Η αυτοραδιογραφική μελέτη των θέσεων δέσμευσης του L-γλουταμικού έδειξε δέσμευση στη κοκκώδη και μοριακή στοιβάδα της ανθρώπινης παρεγκεφαλίδας. Παρουσία ιόντων Cl⁻ παρατηρήθηκε αύξηση στη μοριακή στοιβάδα περίπου 2,5 φορές, ενώ απουσία ιόντων Cl⁻ η δέσμευση ήταν μεγαλύτερη στη κοκκώδη στοιβάδα. 2. Τα πειράματα διαλυτοποίησης έδειξαν ότι το Zwittergent 3-12 είναι το απορρυπαντικό που διαλυτοποιεί σε μεγαλύτερο ποσοστό τις θέσεις δέσμευσης του L-γλουταμικού οξέος σε σχέση με το CHAPS και την διγιτονίνη. 3. Στην ατροφική παρεγκεφαλίδα παρατηρήθηκε μια πολύ σημαντική ελάττωση της ειδικής δέσμευσης του L-[³Η] γλουταμικού οξέος (Bmax) παρουσία ή απουσία ιόντων Cl⁻, ενώ οι τιμές της Kd δεν μετεβλήθησαν. Οι φαρμακολογικές ιδιότητες των θέσεων δέσμευσης του L-[³H] γλουταμικού στην ελαιογεφυροπαρεγκεφαλιδική ατροφία ήσαν παρόμοιες με αυτές της φυσιολογικής παρεγκεφαλίδας.
