A correlation between mast cell population and the degree of inflammations has been suggested. However, comparative studies of mast cell counts in inflammed tissue showed no concurrence. Some investigations have shown an increase of mast cell density which seems to be contradicted by other studies showing a decrease. This study was, undertaken there fore, to investigate the correlation between mast cell number in non insulin dependent diabetic patients (N.I.D.D.M.) and in non diabetic patients.with periodontitis. Subjects for this study were 60 patients. Their ages ranged between 40 - 56 years: 37 of them were non insulin dependent diabetic patients - 23 diabetic patients with periodontitis and with a loss of attachment about 5- 6 mm. and 14 diabetic patients with clinically normal gingiva. Also 23 of them were non diabetic patients presenting no other systematic disorder, 16 of the above were non diabetic patients with periodontitis and with a loss of attachment about 5- 6 m.m. and 7 persons with clinically normal gingiva. The diabetic patients were referred from the Diabetic clinic of Hospital «Evagelismos» and the non diabetic patients were referred from the Periodontology clinic of the Athens University. Biopsy specimens were taken with modified Widman flap which is a technique recognized as well as the open flap curettage technique. Histological slides were prepared for each patient case and stained according to the followingtechniques: a) Hematoxylin - Eosin (H-E) b) Toluidine blue (T.B.) 0,5%. The (H - E) stainwas used to mark the degree of inflammation in O, 1, 2, 3, 4. Quantative estimation of mast sell has been done succesfullyin the histological material stained with 0,5% T.B. The specimens were examined by light microscopy at X 40 and there have been three measuriments of mast cells were taken in three different optical planes. The results of the present study lead to the following conclusions: 1) The mean value of mast cells number is non greater in non insulin dependent diabetic patients with periodontitis than in non diabetic patients with periodontitis. 2) The mean value of mast cells is number is greater in non diabetic patients with periodontitis than in diabetic patients with clinically normal gingival. 3) The mean value of must cells number seems to be independent of the degree of inflammation in diabetic patients as well as in non diabetic patientswith periodontitis. 4) It is not possible to measure the precise number of mast cells by histological techniques, but T.B. 0,5% is a good stain, for the quantative estimation of the number of the mast cells in the gingiva. 5) The shape of mast cells in this study also varies from round to elongated.
