Liquid chromatography-mass spectrometry, an evolution in doping analysis

dc:descriptionBefore the introduction of LC-MS in the field of doping analysis, mass spectrometrical information was acquired with capillary GC-MS. This analytical technique is only suitable for volatile compounds. Because doping agents include polar functional groups in their structure, their volatility is limited and prior to GC-MS analysis, derivatisation is required for most of them. Unfortunately, some derivatives are thermal labile and can not be detected. Liquid chromatography hyphenated with mass spectrometry (LC-MS) does not require a derivatisation step. This analytical technique was introduced in the doping laboratories during the nineties. The first application of LC-MS described in this work was for the detection of the highly polar and thermally labile diuretics. A sensitive screening method for their urinary detection was developed and validated. Further research revealed that also for corticosteroids (non polar doping agents) good results can be achieved with LC-MS. Sensitivity, selectivity and specificity for this group of compounds could not be reached by any other analytical technique. In a next part of this thesis LC-MS was applied for the detection of selected anabolic agents and androst-4-ene-3,617-trione which can act as an aromatase-inhibitor. Finally, the application of LC-MS for the urinary detection of stimulants was investigated.
