Superoxide anions O2- do not exhibit direct apoptosis inducing potential, whereas their dismutataion product hydrogen peroxide H2O2 induces apoptosis nonselectively. Myeloperoxidase MPO converts H2O2 into the selective apoptosis mediator HOCl, which interacts with transformed target cell derived O2- and generates apoptosis inducing hydroxyl radicals OH. Nitric oxide NO does not directly induce apoptosis in fibroblasts. Interaction of NO with transformed cell derived O2- leads to the generation of the apoptosis inducer peroxynitrite ONOO-, which itself does not discriminate between transformed and nontransformed cells. Apoptosis induction by NO as well as the bystandereffect can be blocked by SOD.HOCl does not induce apoptosis in nontransformed fibroblasts, in contrast to their transformed descendants. However, HOCl generation by the combination of glucose oxidase and MPO in the presence of Cu-SOD causes apoptosis induction in nontransformed cells. This effect depends on the concentration of Cu-SOD and is not seen for Mn-SOD, showing the necessity of Cu (a mediator of the Fenton reaction). As heat-inactivated Cu-SOD is not able to induce apoptosis, the Cu-containing functional enzyme seems to be necessary. Cell death induced by Cu-SOD is inhibited by the HOCl-scavenger taurine, the ?OH -scavenger terephthalate and the O2--scavenger Mn-SOD.
