Prognostic markers and molecular characteristics of the tumour suitable to predict the individual course of the disease gain more and more interest due to the possibilities for risk adapted therapy and individualization of follow-up. <br>In addition to the therapeutic consequences drawn from Estrogen (ER)- and Progesteron Receptor (PR)-expression of breast cancer, other members belonging to the superfamily of steroid hormone receptors are still under investigation. Among those are the Vitamin-D3-Receptor (VDR) and the Androgen Receptor (AR) as well as the only recently described tissue-specific cofactor of the AR, the Four-and-a-half-LIM-domain-protein-2 (FHL-2). Furthermore, the Bone Sialoprotein (BSP) might be involved in the skeletal homing of tumour cells. <br>The primary goal of this study was to examine retrospectively the expression of VDR, AR, FHL-2 and BSP in human breast cancer. Therefore, archival samples were submitted to immunohistochemical analysis. As an attempt to correlate individual expression patterns of these proteins with the metastatic behaviour of the tumour, samples were selected according to the clinical outcome. Besides samples derived from patients being free of metastatic disease for at least five years after diagnosis and therapy of the primary tumor (control group, n = 21), we enrolled samples from patients suffering from metastatic disease either at primary diagnosis or during follow up. Samples obtained from patients with systemic disease were further classified into two groups: tumours derived from patients with visceral metastases (group 2, n = 9) and those derived from patients with skeletal manifestation of the disease (group 3, n = 15). <br>All tumours investigated were seen to express VDR, AR, FHL-2 and BSP. However, we were unable to detect any correlation between expression of BSP and/or FHL-2 and metastatic behaviour of the tumour. Furthermore, the recently mentioned correlation between BSP-expression and the occurrence of skeletal metastases was not seen among our patients. This might be due to the rather small number of samples available for this study.
