EHEC-Bakterien setzen Shigatoxine frei, die in vivo und in vitro renale Endo- und Epithelzellen schädigen. <br>Durch Beurteilung der Zellmorphologie, Bestimmung des Hämatoxylin-Labeling Index nach Hämatoxylinfärbung und Annexin-V-Immunfluoreszenzmikroskopie konnte in der vorliegenden Arbeit gezeigt werden, daß sowohl Shigatoxin1 (Stx1) und als auch Shigatoxin2 (Stx2) toxisch auf proximale Tubulusepithelzellen der Niere (LLC-PK1) wirken und konzentrationsabhängig Apoptose auslösen: [Kontrolle: 10%, Stx1 0,1µg/ml: 14% (p>0,05), Stx1 0,5µg/ml: 29% (p<0,001) , Stx1 1,0µg/ml: 37% (p<0,001), Stx2 0,1µg/ ml: 21% (p<0,05), Stx2 0,5µg/ml: 33% (p<0,001), Stx2 1,0µg/ml: 41% (p<0,001)]. Shigatoxin2 besaß dabei ein stärkeres toxisches Potential als Shigatoxin1. Durch fluorometrisch markierte Anti-CD77-Antikörper konnte auf LLC-PK1-Zellen die Expression der funktionellen Shigatoxin- (Gb3-) Rezeptoren nachgewiesen werden. Es ist bekannt, daß TNF-alpha; die Expression von Gb3-Rezeptoren auf Endothelzellen steigert und dadurch die Shigatoxinwirkung verstärkt. Auf LLC-PK1-Zellen (Epithelzellen) dagegen steigerte die Vorbehandlung mit TNF-alpha (10ng/ml) die Gb3-Rezeptorexpression nur geringfügig. TNF-alpha (10ng/ml) löste ebenso wie Shigatoxine Apoptose aus (Kontrolle: 10%, TNF-alpha: 23%, p<0,001). Eine zusätzliche Verstärkung der Shigatoxinwirkung durch TNF-alpha konnte nur für niedrige Shigatoxin1-Konzentrationen (0,1µg/ml) nachgewiesen werden. Die Messung der Caspase-3-Aktivität mit fluorometrisch markiertem Caspase-3-Substrat ergab eine Aktivierung der „Effektor“-Caspase-3 durch Shigatoxin1 und 2. Nach Inkubation mit beiden Shigatoxinen führte eine gleichzeitige Behandlung mit Disulfiram (Tetraethylthiuram Disulfid), einem konzentrationsabhängigen Inhibitor der Caspase-3, in LLC-PK1-Zellen zu einem signifikant geringeren Anteil apoptotischer Zellen (Stx1: -19%, p<0,001 bzw. HUS is predominantly associated with Enterohemorrhagic Escherichia coli (EHEC) infection. In this context, the release of shigatoxins (Stx) by EHEC is thought to be the major pathogenic mechanism. <br>The aim of our study was to get more information about the role of Stx and Tumor Necrosis Factor-alpha (TNF-alpha) in the pathophysiology of HUS and to define more precisely the mode of Shigatoxin-induced cell death with special regard to the path by which cell death/apoptosis is generated. <br>In LLC-PK1-cells, a cell line from the kidney of a Hampshire pig, that is well established and generally accepted as a model of proximal tubular epithelial cells, Stx 1 and Stx 2 caused apoptosis in a concentration dependent manner, as demostrated by analysis of cell-morphology, Annexin-V-immunfluorescence-microscopy and Hematoxylin-Staining. In this context, Stx2 was shown to be more toxic than Stx 1. <br>Occurrence of Gb3-receptors, the functional receptors for Stx, on the surface of LLC-PK1-cells, has been shown by Anti-CD77-immunfluorescencemicroscopy. <br>In endothelial cells, pre-incubation with TNF-alpha increased Stx-induced apoptosis through an increased density of Gb3–receptors on the cell surface. In contrast, in LLC-PK1-cells (epithelial cells) incubation with TNF-alpha induced apoptosis, slightly increased the number of Gb3-receptors, and boosted Stx-induced apoptosis at low concentrations of Stx1 only. <br>In our cell model Stx 1 and Stx 2 caused activation of effective caspase-3, as indicated by the fluorometric PhiPhiLux-Caspase-3-activity assay. As demostrated by Annexin-V-immunfluorescence and Hematoxylin-Staining in LLC-PK1-cells, apoptosis induced by exposure to Stx 1 and Stx2 could be inhibited by incubation with Tetraethyldisulfid (TED, Disulfiram).
