Die Wirkung von VEGF wird über spezifische Rezeptoren vermittelt, die zur Familie der Rezeptor-Tyrosinkinasen gehören. Ein neues Konzept in der Therapie der angioproliferativen Retinopathie könnte die Blockade dieser Rezeptoren darstellen, um den Effekt von VEGF abzuschwächen oder gar aufzuheben. <br>Ziel dieser Arbeit war es, die Wirkung einer lokalen Therapie durch intravitreale Injektion von VEGF-Rezeptorblockern auf die Ausprägung der experimentell erzeugten Sauerstoff-induzierten angioproliferativen Retinopathie (OIR) im Mausmodell nach Smith zu untersuchen. Zum Einsatz kamen die drei Substanzen PTK787, MAE87 und MAZ186, die zur Gruppe der \"small molecule receptor tyrosine kinase inhibitors\" gehören. Das retinale Gefäßsystem wurde in Netzhautflachpräparaten nach Perfusion mit hochmolekularem Flourescein-gekoppeltem Dextran dargestellt. Anhand eines von Higgins erstellten und von uns modifizierten Auswertungsschemas wurde im intraindividuellen Vergleich zwischen behandeltem Auge und Kontrollauge der Therapieerfolg ermittelt. <br>Die Behandlung mit PTK787 (1x 40 mM) und MAE87 ergab jeweils eine hochsignifikante Reduktion der Ausprägung der OIR (p < 0,01). Dagegen konnte mit MAZ186 und PTK787 (5 mM bzw. 2x 40 mM) kein signifikantes Ergebnis erzielt werden. The effect of VEGF is mediated by specific receptors which belong to the family of tyrosine kinase receptors. Inhibition of these receptors in order to reduce the effect of VEGF could be a new concept in the therapy of angioproliferative retinopathy. It was the aim of this work to examine the effect of a local therapy by intravitreal injection of VEGF-receptor inhibitors on oxygen-induced angioproliferative retinopathy (OIR) in mice. Three different substances were tested: PTK787, MAE87 and MAZ186. They all belong to the class of \"small molecule receptor tyrosine kinase inhibitors\". The vascular system was visualised in retinal whole mounts by intracardial perfusion of high molecular flourescein-labeled dextran. We modified a retinopathy score developed by Higgins et al. which measures the angioproliferative changes and compared intraindividually the scores of drug injected eye and control eye. <br>The injection of PTK787 (1x 40 mM) and MAE87 reduced the severeness of the OIR highly significant (paired t-test: p < 0,01). The treatment with MAZ186 and PTK787 (5 mM bzw. 2x 40 mM) on the other side did not result in significant changes. None of the three tested substances had a negative effect on the fully developed murine retinal vascular system.
