Analytical methods for the determination of endocrine disruptors, UV filters and plastic additives in packaging and pharmaceuticals

This thesis is presented as a compilation of publications and deals with the development, optimization and validation of different analytical methods to determine in packaging: UV filters by HPLC-DAD, plastic additives by UHPLC-(ToF)MS and perfluorinated compounds by UHPLC-(QToF)MS/MS. In each work, a focused ultrasonic solid-liquid extraction (FUSLE), a fast, simple and relatively new extraction technique, is developed and optimized; and its efficiency is evaluated by comparing it with other extraction techniques. Lastly, the developed methods are applied to the analysis of several real samples. In the case of the UV filters method, it was also applied to evaluate the migration and delamination capacities of these analytes through different contact layers in multilayer packaging. Finally, on a line parallel to these works, an analytical method for the determination of parabens in pharmaceuticals by SLE-SPE-UHPLC-(QqQ)MS/MS was developed. Moreover, the optimized method was also applied to the analysis of more than one hundred samples in order to evaluate if the use of medicines may be a source of exposure to parabens. Esta tesis se presenta como compendio de publicaciones y trata del desarrollo, optimización y validación de diferentes métodos de análisis para la determinación en el embalaje de filtros UV por HPLC-DAD, aditivos plásticos por UHPLC-(ToF)MS y compuestos perfluorados por UHPLC-(QToF)MS/MS. En cada trabajo, una extracción sólido-líquido con ultrasonidos focalizados (FUSLE), una técnica de extracción rápida, sencilla y relativamente nueva, es desarrollada y optimizada. Asimismo, la eficacia del método FUSLE es evaluada comparándola con otras técnicas de extracción. Finalmente los métodos desarrollados se aplican para el análisis de varias muestras reales. En el caso del método para determinar filtros UV, también se aplicó para estudiar los procesos de migración y delaminación de los filtros UV en el embalaje flexible multicapa a través de diferentes capas de contacto. Finalmente, en una línea paralela, se desarrolló un método analítico para la determinación de parabenos en productos farmacéuticos por SLE-SPE-UHPLC-(QqQ)MS/MS. Además, el método desarrollado se aplicó para el análisis de más de cien muestras con el fin de evaluar si el consumo de medicamentos puede ser una fuente de exposición a los parabenos.
