Using the method of layer analysis and radioactive tracers the diffusion coefficients and self diffusion coefficients of several antibiotics and of glucose, sucrose, inulin and decamethonium iodide in mucus have been determined. Human bronchial mucus subjected to a homogenization - reconstitution process was used to overcome problems associated with the known heterogeneity of this material. The results have been discussed with respect to current theories of diffusion of solutes in gels. It has been shown that the only barrier to the diffusion of the studied solutes in mucus arises from an obstructional effect due to the gel matrix material. It was found that a linear relationship existed between the diffusion coefficients of the solutes and the percentage dry weight of the mucus gel. The possibility of binding of solutes by bronchial mucus has also been investigated. Using the techniques of ultrafiltration and equilibrium dialysis the binding of benzyl-penicillin, cephalexin, chloramphenicol. tetracycline and decamethonium iodide to reconstituted human bronchial mucus and to human serum albumin has been determined. lt was found that only tetracycline was bound to any appreciable degree by bronchial mucus whereas all the antibiotics were bound to serum albumin. The results were analysed and where possible binding parameters have been calculated and possible mechanisms of binding discussed.
