The ability of the transformants/transductants to grow on succinate as the sole non-fermentable carbon source was used to select mutants with a functional F₁F₀ ATPsynthase. Many F₁ -complementing plasmids and cosmids were isolated from the four E. coli F₁ unc point mutants screened.
