Facing the problem of MPs accumulation that will undoubtedly cause leaks clogging over time and that are still no proven methods by the scientific community, several harmonious procedures have been proposed to determine MPs in any type of matrix. This study presents a scoop of MPs extraction in dry-weighed sludge samples. Given a variety of protocols, differing retention times and digestion efficacy have been compared, using the principle of density separation followed by filtration with size fractioning, the results in each protocol have been different. Four protocols were intercompared to assert the major and apply it on a real mud. For the validation of the method, 30 g of sludge was used (ielab 0301-Spain) with 2 types of polyolefins and nylon. The sludge had undergone a division into 2 fractions according to four protocols, each protocol running triplicates and blanks. The first Hcl-based protocol was inspired by Cole et al., (2014) study. The second protocol of n-hexane. Instead of Hcl, the recommendation of Shim et al., (2016) to use n-hexane was followed to increase the potential for comparability. The third H2O2 enzyme two-step protocol was an alternative, stable against most of the mild chemical existing treatments. Two portions of H2O2 were used given its property of bleaching while rational heating, according to Sujathan et al., (2017). The last protocol was an enhanced version of Löder et al., (2017) study. Data collection depicted by an optical visualization and chemical identification, via microscopy and Infrared Spectroscopy (FT-IR) to record polymer spectra was established to interpret the potential retrieval. The results revealed that MP recuperation with the third protocol based on hemicellulase and H2O2 was more efficient, there was no large fluctuation in MP number nor interference caused by the reaction reagents. Then, the optimal extraction method for MPs in sewage sludge is suggested to be digested with hemicellulase at 50 ºC, followed by bleaching with H2O2. Conclusively, in the short run, it is recommended to optimize this protocol to claim a faster and more economical operational system while seeking to increase MPs' recovery rates. Moreover, a higher quantity of MPs in real sludges was found, ranging from 96 to 769 particles/g dry sludge (ds), with an average of 392.4 ± 72.01 particles/g ds, with the dominance of 100-355 m sizes, commonly fibres. Thermoplastics, ethylene derivatives, polyethylene terephthalate and polyamide were the dominant MPs. The estimation of MPs for the province of Cádiz (2.34 × 108 MPs/tonne) made it possible to estimate an average number of MPs for Spain at 2.80 × 1014 MPs per year, which can go to other terrestrial environmental compartments through the sludge generated by the WWTPs of the country. To know the possible impact of MPs on the use of sludge as agricultural fertilization, the presence of MPs in agricultural soils previously treated with sludge or compost from sewage treatment plants for fertilization, and their exposure to earthworms in southern Spain was evaluated. Composts were used, selected at different stages of the composting process (humid, semi-dried and fine composts), and different types of fertilised soils at ascending doses (S1, S2 and S3), taking samples at different depths (0-5, 5-10 and 10-20 cm) to study the shape, size, type and abundance of MPs using FT-IR. Two earthworm species have been also studied as bioindicators to check the level of ingestion at increasing doses of the common MPs encountered (0.9, 2.1 and 3% w/w). Research on MPs contamination in soils was not either surprising, as soils are a grave of fragmented agricultural mulch film, organic fertilizer, sewage irrigation, sludge/compost amendment, atmospheric deposition and surface runoff. So far, results indicated the presence of higher amounts of 408.3 ± 12.4 × 106 and 176.2 ± 15.5 × 106 MPs/tonne, in studied composts and soils respectively. The distribution of MPs types, most of them were fibres, differentiated by size and types (polytetrafluoroethylene, thermoplastic polyamide elastomer, polypropylene, polyvinyl chloride and polyethylene terephthalate) evolved differently, the predominant size was 250 m. Continuously, and in order to demonstrate the transition of the micropollutants to the biota, the results revealed significant ingestion of MPs in earthworms with increasing doses of the common MPs encountered ranging from 2.1 to 3% w/w, characterized mostly by polypropylene, as the most ingested polymer type. From a future perspective, the research topic highlights one of the most important problems at present, such as the contamination of MPs from daily use and the lives of people, which reach the terrestrial environment through the sludge of the treatment plants The utility of this doctoral thesis is twofold: on the one hand, it sheds light so that future regulations (both for wastewater treatment and the application of sewage sludge) take into account the need to control this pollutant, which has come to stay. On the other hand, it reveals the impact that it may be generating for decades and unknowingly on the terrestrial microbiota. La alarma ambiental global y la sensibilidad social revelaron que los microplásticos (MPs) son el tipo más notable de degradación y contaminación plástica que ha llamado mucho la atención entre los investigadores, quienes han detectado rastros de MPs en ambientes marinos y terrestres. Ante el problema de la acumulación de MPs que indudablemente provocará la obstrucción de fugas con el tiempo, se han propuesto varios procedimientos armoniosos para determinar MPs en cualquier tipo de matriz. Este estudio presenta una primicia de extracción de MPs en muestras de lodos pesados en seco. Dada una variedad de protocolos, se han comparado diferentes tiempos de retención y eficacia de la digestión, utilizando el principio de separación por densidad seguida de filtración con fraccionamiento por tamaño, los resultados en cada protocolo han sido diferentes. Se intercompararon cuatro protocolos para optimizar el mejor y aplicarlo sobre un lodo real. Para la validación del método, se emplearon 30 g de lodo de referencia ielab 0301-España con 2 tipos de poliolefinas y nylon. El lodo había sufrido una división en 2 fracciones de acuerdo con cuatro protocolos, cada protocolo ejecutando triplicados y controles. El primer protocolo basado en Hcl se inspiró en el estudio de Cole et al., (2014). El segundo protocolo de n-hexano. en lugar de Hcl, según la recomendación de Shim et al., (2016) de usar n-hexano para aumentar el potencial de comparabilidad. El tercer protocolo de dos fases de H2O2 - enzima fue una alternativa estable frente a la mayoría de los tratamientos químicos suaves existentes. Se utilizaron dos porciones de H2O2 dada su propiedad de blanquear mientras se calienta, según Sujathan et al., (2017). El último protocolo fue una versión mejorada del estudio de Löder et al., (2017), se estableció la recopilación de datos representados por una visualización óptica e identificación química a través de microscopía y espectroscopía infrarroja (FT-IR) para registrar espectros de polímeros e interpretar la recuperación potencial. Los resultados revelaron que la recuperación de MPs con el tercer protocolo basado en hemicelulasa y H2O2 fue más eficiente, no hubo una gran fluctuación en el número de MPs ni interferencias causadas por los reactivos de reacción. Luego, se sugiere que el método óptimo de extracción de MPs en lodos de depuradora sea digerir con hemicelulasa a 50 ºC, seguido de blanqueo con H2O2. En conclusión, a corto plazo, se recomienda optimizar este protocolo para reclamar un sistema operativo más rápido y económico mientras se busca aumentar las tasas de recuperación de los MPs. Además, se encontró una mayor cantidad de MPs en los lodos reales, que van desde 96 a 769 partículas/g lodo seco (ps, peso seco), con un promedio de 392,4 ± 72,01 partículas/g ps, y con predominio de tamaños de 100-355 μm, comúnmente fibras. Los termoplásticos, los derivados del etileno, el tereftalato de polietileno y la poliamida fueron los MPs dominantes. La estimación de MPs para la provincia de Cádiz (2.34 × 108 MPs/tonelada) permitió estimar una cantidad promedio de MPs para España en 2,80 × 1014 MPs al año, que pueden ir a otros compartimentos ambientales terrestres a través de los lodos generados por las EDARs del país. Para conocer el posible impacto por MPs del uso de lodos como fertilización agrícola, se evaluó la presencia de MPs en suelos agrícolas previamente tratados con lodos o composta de depuradoras para su fertilización, y su exposición a las lombrices en el sur de España. Se utilizaron compostas seleccionadas en diferentes etapas del proceso de compostaje (compostas húmedas, semisecas y finas), y diferentes tipos de suelos fertilizados en dosis ascendentes (S1, S2 y S3), tomando muestras a diferentes profundidades (0-5, 5-10 y 10-20 cm) para estudiar la forma, tamaño, tipo y abundancia de MPs usando FT-IR. También se han estudiado dos especies de lombrices de tierra como bioindicadores para comprobar el nivel de ingestión en dosis crecientes de los MPs comunes encontrados (0,9, 2,1 y 3 % p/p). La investigación sobre la contaminación de MPs en los suelos tampoco fue sorprendente, ya que los suelos son una tumba de películas de mantillo agrícola fragmentada, fertilizantes orgánicos, riego de aguas residuales, enmienda de lodo/composta, deposición atmosférica y escorrentía superficial. Hasta el momento, los resultados indicaron la presencia de cantidades mayores de 408.3 ± 12.4 × 106 y 176.2 ± 15.5 × 106 MPs/tonelada, en compostas y suelos estudiados respectivamente. La distribución de los tipos de MPs, la mayoría de ellos de fibra, diferenciados por tamaño y tipos (politetrafluoroetileno, elastómero de poliamida termoplástica, polipropileno, cloruro de polivinilo y tereftalato de polietileno) evolucionó de manera diferente, el tamaño predominante fue el de 250 μm. Continuamente, y con el fin de demostrar la transición de los microcontaminantes a la biota, los resultados revelaron una ingestión significativa de MPs en las lombrices de tierra con dosis crecientes de los MPs comunes encontrados que oscilaban entre 2,1 y 3 % p/p, y el polipropileno fue el tipo de MPs más ingerido. Desde las perspectivas futuras, el tema de investigación pone de relieve uno de los problemas más importantes en la actualidad como es la contaminación de MPs procedentes del uso cotidiano y de la vida de las personas, que llegan al medio terrestre a través de los lodos de las depuradoras. La utilidad de esta tesis doctoral es doble: por un lado, aporta luz para que las futuras normativas (tanto de depuración de aguas residuales, como de aplicación de lodos de depuradora) tengan en cuenta la necesidad de controlar este contaminante, que ha venido para quedarse. Por otro lado, pone de manifiesto el impacto que puede estar generando desde décadas y sin saberlo sobre la microbiota terrestre. Desde mi punto de vista, se recomiendan más ensayos avanzados de toxicidad que arrojen conocimiento sobre el impacto que pueda tener, en este caso sobre las lombrices de tierra.","L'utilisation intensive du plastique vise à rendre la vie plus facile, avec la valeur la plus élevée et l'apport le plus faible en ressources. L'alarme environnementale mondiale et la sensibilité sociale ont révélé que les microplastiques (MPs) sont le type le plus notable de dégradation et de pollution plastique qui a attiré beaucoup d'attention parmi les chercheurs, qui ont détecté des traces de MPs dans les environnements marins et terrestres. Face au problème de l'accumulation des MPs qui causera sans aucun doute le colmatage des fuites au fil du temps et qui n'a pas encore fait l'objet de méthodes éprouvées par la communauté scientifique, plusieurs procédures harmonieuses ont été proposées pour déterminer les MPs dans n'importe quel type de matrice. Cette étude présente un scoop d'extraction des MPs dans des échantillons de boues pesées sèches. Plusieurs protocoles ont été comparés, avec des temps de rétention et des efficacités de digestion différents, en utilisant le principe de la séparation par densité suivie d'une filtration avec fractionnement de la taille, les résultats de chaque protocole étant différents. Quatre protocoles ont été comparés entre eux afin d'affirmer la méthode principale et de l'appliquer à une boue réelle. Pour la validation de la méthode, 30 g de boue ont été utilisés (ielab 0301-Espagne) avec 2 types de polyoléfines et du nylon. La boue a été divisée en deux fractions selon quatre protocoles, chaque protocole comportant des triplicats et des blancs. Deux portions de H2O2 ont été utilisées en raison de sa propriété de blanchir tout en chauffant rationnellement, selon Sujathan et al., (2017). Le dernier protocole était une version améliorée de l'étude de Löder et al., (2017). La collecte de données décrite par une visualisation optique et une identification chimique, via la microscopie et la spectroscopie infrarouge (FT-IR) pour enregistrer les spectres de polymères a été établie pour interpréter la récupération potentielle. Les résultats ont révélé que la récupération des MPs avec le troisième protocole basé sur l'hémicellulase et le H2O2 était plus efficace, qu'il n'y avait pas de fluctuation importante du nombre de MPs ni d'interférence causée par les réactifs de réaction. Il est donc suggéré que la méthode optimale d'extraction des MPs dans les boues d'épuration soit la digestion par l'hémicellulase à 50 ºC, suivie d'un blanchiment au H2O2. En conclusion, à court terme, il est recommandé d'optimiser ce protocole pour prétendre à un système opérationnel plus rapide et plus économique tout en cherchant à augmenter les taux de récupération des MPs. En outre, une plus grande quantité de MPs dans les boues réelles a été trouvée, allant de 96 à 769 particules/g de boue sèche (ds), avec une moyenne de 392,4 ± 72,01 particules/g ds et avec la dominance des tailles de 100-355 μm, généralement des fibres. Les thermoplastiques, les dérivés de l'éthylène, le polyéthylène téréphtalate et le polyamide étaient les MPs dominants. L'estimation des MPs pour la province de Cadix (2,34 × 108 MPs/tonne) a permis d'estimer le nombre moyen de MPs pour l'Espagne à 2,80 × 1014 MPs par an, qui peuvent se retrouver dans d'autres compartiments de l'environnement terrestre, par le biais des boues générées par les stations d'épuration du pays. L'utilité de cette thèse de doctorat est double : d'une part, elle éclaire pour que les réglementations futures (tant pour le traitement des eaux usées que pour l'application des boues d'épuration) tiennent compte de la nécessité de contrôler ce polluant, qui s'est imposé. Global environmental alarm and social sensitivity revealed that microplastics (MPs) are the most notable type of plastic degradation and pollution that has attracted much attention among researchers, who have detected MP traces in marine and terrestrial environments. Facing the problem of MPs accumulation that will undoubtedly cause leaks clogging over time and that are still no proven methods by the scientific community, several harmonious procedures have been proposed to determine MPs in any type of matrix. This study presents a scoop of MPs extraction in dry-weighed sludge samples. Given a variety of protocols, differing retention times and digestion efficacy have been compared, using the principle of density separation followed by filtration with size fractioning, the results in each protocol have been different. Four protocols were intercompared to assert the major and apply it on a real mud. For the validation of the method, 30 g of sludge was used (ielab 0301-Spain) with 2 types of polyolefins and nylon. The sludge had undergone a division into 2 fractions according to four protocols, each protocol running triplicates and blanks. The first Hcl-based protocol was inspired by Cole et al., (2014) study. The second protocol of n-hexane. Instead of Hcl, the recommendation of Shim et al., (2016) to use n-hexane was followed to increase the potential for comparability. The third H2O2 enzyme two-step protocol was an alternative, stable against most of the mild chemical existing treatments. Two portions of H2O2 were used given its property of bleaching while rational heating, according to Sujathan et al., (2017). The last protocol was an enhanced version of Löder et al., (2017) study. Data collection depicted by an optical visualization and chemical identification, via microscopy and Infrared Spectroscopy (FT-IR) to record polymer spectra was established to interpret the potential retrieval. The results revealed that MP recuperation with the third protocol based on hemicellulase and H2O2 was more efficient, there was no large fluctuation in MP number nor interference caused by the reaction reagents. Then, the optimal extraction method for MPs in sewage sludge is suggested to be digested with hemicellulase at 50 ºC, followed by bleaching with H2O2. Conclusively, in the short run, it is recommended to optimize this protocol to claim a faster and more economical operational system while seeking to increase MPs' recovery rates. Moreover, a higher quantity of MPs in real sludges was found, ranging from 96 to 769 particles/g dry sludge (ds), with an average of 392.4 ± 72.01 particles/g ds, with the dominance of 100-355 m sizes, commonly fibres. Thermoplastics, ethylene derivatives, polyethylene terephthalate and polyamide were the dominant MPs. The estimation of MPs for the province of Cádiz (2.34 × 108 MPs/tonne) made it possible to estimate an average number of MPs for Spain at 2.80 × 1014 MPs per year, which can go to other terrestrial environmental compartments through the sludge generated by the WWTPs of the country. To know the possible impact of MPs on the use of sludge as agricultural fertilization, the presence of MPs in agricultural soils previously treated with sludge or compost from sewage treatment plants for fertilization, and their exposure to earthworms in southern Spain was evaluated. Composts were used, selected at different stages of the composting process (humid, semi-dried and fine composts), and different types of fertilised soils at ascending doses (S1, S2 and S3), taking samples at different depths (0-5, 5-10 and 10-20 cm) to study the shape, size, type and abundance of MPs using FT-IR. Two earthworm species have been also studied as bioindicators to check the level of ingestion at increasing doses of the common MPs encountered (0.9, 2.1 and 3% w/w). Research on MPs contamination in soils was not either surprising, as soils are a grave of fragmented agricultural mulch film, organic fertilizer, sewage irrigation, sludge/compost amendment, atmospheric deposition and surface runoff. La alarma ambiental global y la sensibilidad social revelaron que los microplásticos (MPs) son el tipo más notable de degradación y contaminación plástica que ha llamado mucho la atención entre los investigadores, quienes han detectado rastros de MPs en ambientes marinos y terrestres. Ante el problema de la acumulación de MPs que indudablemente provocará la obstrucción de fugas con el tiempo, se han propuesto varios procedimientos armoniosos para determinar MPs en cualquier tipo de matriz. Este estudio presenta una primicia de extracción de MPs en muestras de lodos pesados en seco. Dada una variedad de protocolos, se han comparado diferentes tiempos de retención y eficacia de la digestión, utilizando el principio de separación por densidad seguida de filtración con fraccionamiento por tamaño, los resultados en cada protocolo han sido diferentes. Se intercompararon cuatro protocolos para optimizar el mejor y aplicarlo sobre un lodo real.
