It not only affects communication, but also leads to other disorders, such as tinnitus and hyperacusis. Tinnitus, ringing in the ear, is a devastating disorder affecting millions, especially those who have hearing loss. Tinnitus is believed to be caused by plasticity changes in the central auditory system (CAS) after hearing loss. A common symptom associated with tinnitus is hyperacusis. Hyperacusis defined as hypersensitivity to normal environmental sounds, has a prevalence in the tinnitus population estimated to be as high as 80%. Some reports suggest that hyperacusis is linked to childhood hearing loss. To date, the neural mechanisms of hyperacusis are largely unknown, and there are no widely accepted treatments to cure this disorder. A major aim of this dissertation is to study the effect of early-age noise exposure on sound loudness perception. I used high-level and low-level noise exposure to explore the effect of noise exposure at early-age in changing the sound loudness sensitivity in later adulthood. I found that any auditory disturbance in the auditory system during the developmental period could be a risk factor for loudness changes; an indication of hyperacusis. A second aim of this dissertation is to investigate the neural sources of hyperacusis via measuring the neural activity in the auditory structures, such as the auditory cortex (AC) and the inferior colliculus (IC), and in the limbic system, such as the nucleus accumbens (NAc). Hyperexcitability in the AC, IC and NAc was examined using c-Fos staining. Increased c-Fos staining was evident in the IC and the NAc post low-level noise exposure. These results suggest that low-level noise, even though it may not cause permanent hearing loss at early-age may still be a risk factor for hyperacusis. Reduced c-Fos staining was detected post-high-level noise exposure presumably due to hearing loss.
