A whole new idea on the modification of the restorative resins currently used for caries treatment was first hypothesized by employing polymer-drug conjugate (PDC), more specifically polymer-antibiotic conjugate (PAC), which makes the resin not only a filling material but an antibiotic delivery system which, in principle, could ensure antimicrobial effectiveness for a much longer time period as compared with directly using small-molecular antibiotics as additives. Moreover, the polymer backbone of PAC will be constructed using a commonly used monomer in the dental resin, i.e. 2-hydroxyethyl methacrylate (HEMA), to promote compatibility between the resin and the PAC during the mixing and further curing process. Thus this thesis aims to provide a general strategy for the synthesis of these PACs and PAC-added dental resins, and to demonstrate physical properties and antimicrobial performances of these new resins using the blank resin and the antibiotic as controls, respectively. In this proof-of-concept study, ciprofloxacin was selected as the antibiotic because it is very commonly used and has multiple functional group for modification. The preparation and characterization of ciprofloxacin-based monomer-antibiotic-conjugate and the corresponding PACs will be addressed in this thesis.
