Antigen analysis of the genus Candida

The purpose of the investigation was to apply the Ouchterlony Technique as a method of immunological assay of several species of Candida. The diffusion of antigen and antibody through the agar resulted in the formation of specific precipitates. The antisera were prepared by immunizing rabbits with formalin-killed suspensions of Candida sp. organisms. Heat and trypsin extracts of Q. guillermondi, Q. parakrusei, c. stellatoidea, Q. tropicalis, C. pseudotropicalis, C. al bicans, C. krusei , and Q. brumpti were used as the source of antigen. Preliminary investigations were done to indicate which extract and dilution gave the best results when tested with whole homologous and heterologous antisera. The observed reactions indicated that the undiluted heat extracts produced better zones of precipitation; these extracts were then tested against concentrated homologus and heterologous immunesera.
