Adequação da metodologia do teste de tetrazólio para sementes de Aspidosperma pyrifolium mart

dc:description.abstractMany forest species are less known according to physiological quality of its seeds, these information are viable, as well as its own preservation or to other means. This work had the aim the adaptation of methodology to elaborate a protocol to execute tetrazolium test on seeds of pereiro – Aspidosperma pyrifolium Mart. Seeds came from São João do Cariri (PB) and handly beneficit, afterwards cut laterably and conditioning, for 3 hours, with watered paper sheet 2,5 times its weight at 25 ºC. Seed tegument was left out and embryo subjected to tetrazolium solution, to coloring, on concentrations at 0,025; 0,050; 0,075 e 0,1% during 30, 60, 90 and 120 minutes, on darkness in B.O.D. at 40ºC. After longitudinal separation among cotiledon and embryo axe center, embryo were evaluated according to its uniformity and coloring intensity showed by tissues, being classified in: non-colored and colored with weak, adequate or strong intensities. The results were compared with germination test (conducted in 4 repetitions of 25 seeds at 25 ºC constantly, with a paper roll as substrate) and emergence (conducted in 4 repetitions of 25 seeds, seeding in plastic plates with washed sand in a greenhouse). Based on results, it had been observed that tetrazolium test is efficient to evaluate pereiro, seeds viability, using tetrazolium solution at 0,075%, during 90 minutes, at 40 ºC. Keywords: coloring,
