ESTUDO COMPARATIVO DO EFEITO DA AMBIENTAÇÃO E DOS ANTICOAGULANTES EDTA E CITRATO DE SÓDIO SOBRE A AGREGAÇÃO PLAQUETÁRIA EM AMOSTRAS SANGUÍNEAS DE GATOS DOMÉSTICOS
Abstract
dc:description.abstractThe purpose of the research was to analise the aclimatation and two anticoagulants effects on platelet aggregation and compare the manual and automatic platelet counting methods in blood samples of cats. One group of ten animals was submitted to a pre blood collection aclimatation under the effects of a facial pheromone difusor, one hour long. The other group of tem cats either, did was not submitted to the pre-collection aclimatation. In both groups was collected 1 mL of blood from the saphenous vein with a n.23 scalp and 3 mL syringe. Half volume was transfered to an eppendorf container with EDTA and the other half was transfered to an eppendorf container with sodium citrate 3.2%. The samples was processed on the Clinical Pathology Laboratory of the UFPB Veterinary Hospital, by automatic counter and then the blood smears was prepared. The platelet aggregations was subjectively classified according to the amount of clumps on the lateral margins and feathered edge as discreet, moderate and intense. Only the samples with discreet and moderate aggregation was submitted to the manual counting in Neubauer chamber and blood smear estimation. The results shown that there was no significant statistical difference in the platelet counts between the aclimated and the non aclimated groups. There was significant difference between the blood smear counting and the automatic counting and between the anticoagulants EDTA and sodium citrate 3.2%. The sodium citrate 3.8% was largely responsible for the intense platelet aggregations.
