Chronic myeloid leukemia (CML) is a clonal hematopoietic stem cell disorder characterized by a translocation between the long arms of chromosomes 9 and 22. The translocation of these chromosomes results in the juxtaposition ABL oncogene on chromosome 9 wit h the BRC housekeeping gene of chromosome 22 producing the BCR - ABL1 fusion gene. This fusion gene is transcribed and translated into protein BCR - ABL1. The tyrosine kinase inhibitors (TKIs) act by blocking the binding of ATP at the ABL1 kinase domain inhibi ts phosphorylation and resulting in cell death. Imatinib was the first inhibitor of protein tyrosine kinase BCR / ABL1 to be used and is considered the "gold standard" treatment for patients with newly diagnosed chronic myeloid leukemia. However, some pati ents can develop resistance to imatinib due to mutations in BRC / ABL1 gene. For this reason, other therapies should be evaluated, including treatment with tyrosine kinase inhibitors second generation, dasatinib, and nilotinib. In this study, the recent li terature data were analyzed four years related adverse events associated with imatinib, dasatinib, and nilotinib. They were found 152 articles, of which 24 were used international articles are available online in the LILACS, IBECS, MEDLINE, Cochrane Librar y, and SciELO. Adverse events were seen at all related tyrosine kinase inhibitors investigated. Among the main events are reported the occurrence of pleural effusion from patients treated with dasatinib, rashes during treatment with nilotinib, and edema, a nd gastrointestinal discomfort reported in patients treated with imatinib. For choosing the most appropriate pharmacotherapy for each patient, it is necessary to consider the occurrence of these adverse events, especially for patients with comorbidities su ch as diabetes mellitus, cardiovascular or respiratory problems, as they are risk factors when combined with use of some tyrosine inhibitors kinase.
