Several microscopes to improve lateral resolution in 1D and 2D have been developed by this thesis. It could be shown that an improvement of the optical resolution by a factor of two can be achieved by the new technique developed here by generating a lateral structure within the plane of the object with a freely selectable fringe distance as well as angular directions. It has been shown that excitation by total internal reflection is also possible using this construction. To characterize the instrument extensive physical measurements on beads as well as simulations were carried out. Furthermore, first succesful measurements on biological objects were made. The new microscope offering structured illumination is especially suitable for biological applications since horizontally orientated slides can be observed from below.
