Diabetik ovariektomize sıçanlarda oluşan vasküler disfonksiyonda östrojen reseptörlerinin rolü ve östrojen tedavisinin etkisi

dc:description.abstractEstrogen has protective effects against cardiovascular disease in pre-menopausal women, are decreased in diabetes and menopause. ER? and ERß mediate estrogen effects, but acute effects on vasculature are not completely understood. In this study, we investigated the contribution of estrogen receptors (ERs) ? and ß to vasorelaxation in thoracic aorta rings from control (C), diabetic (D), ovariectomized (O) and ovariectomized-diabetic (OD) rats. The experimental duration was in 8-16 weeks. On the aortic rings with intact endothelium 8 weeks experimental groups after precontraction with phenylephrine (Phe) were obtained direct relaxant effects of the selective ER agonists PPT, DPN and the nonselective ER agonist 17ß-estradiol (E2)., In Control group PPT made more relaxation than natural ligand that effect both of subtypes and selective ERß agonist DPN that results were diminished in O, D, and OD groups. All agonists of ER made in same ratio relaxation in OE and ODE groups. On mesenteric artery 16 weeks of experimental groups obtained Phe dose responsiveness. Dose responses of Phe in D, OE and ODE groups were shifted to the left side in copmparision to Control group. Dose responsiveness of Phe in Ovariectomized group wasn?t different significantly than in Control group. Shifted to the right side the dose responsiveness of Phe in OD group to comparision in Control group but differencess didn?t significant. Mesenteric arteries were incubated with 3 different ER agonists for 15 minutes and then investigated the contractile dose responsiveness of Phe in existence of ER agonists. In C, O, and OE groups ER? agonist PPT and natural ligand 17ß estradiol with similarity shifted dose responses of Phe slope to the right side however ERß agonist DPN didn?t made any changes. In groups D and OD, slopes of Phe in existence of PPT and 17ß estradiol are shifted less than in Control group but DPN didn?t made any differencess. Results of Phe in existence PPT and 17ß estradiol didn?t change in OE and ODE groups but DPN shifted dose response slope to the right side. In our study ER?s agonists decrease effects of Phe that a contraction agency of vessel. ER? agonist PPT has a superior role to comparision with 17ß estradiol and DPN on the aorta and mesenteric artery. Chronical treatment of menopause and diabetes with estrogen masked ER?s espesually ER??s acute positive effects on vessel tonus.Key words: Aorta, Diabetes, Estrogen receptor ? ß (ER?)
