The effect of 7 different salt doses (0, 3, 6, 9, 12, 15 and 18 g/l) on callus induction and plant regeneration in the above-mentioned varieties was investigated through callus culture. In the in vivo stage, such parameters as the speed and vigor of germination, the lengths of shoots and roots, the wet and dry weights and the total water contents were examined through classical germination tests. The data obtained from both studies were compared and the tolerances of the varieties to the salinity were determined.In addition, the common or different gliadin bands in these varieties were established by examining endosperm specific for storage proteins of the varieties with the method of acid-polyacrylamide gel electrophoresis (A-PAGE).As a result of the statistical assesments of the findings, it has been found of that Bezostaja-1, Demir-2000, İkizce-96, Kate-A-1, Kıraç-66 and Pehlivan common wheat varieties and Mirzabey-2000 and Selçuklu-97 durum wheat varieties are more salt tolerant than the other varieties and that more detailed results have been obtained according to the in vitro methods than in vivo methods which have been used.","abstract_html":"This research, in which mature embryos of 28 common and 13 durum wheat varieties, all of which are registered and currently grown in Turkey, were used as plant materials, was carried out in Biotechnology Laboratory, Department of Field Crops, Faculty of Agriculture in Ankara University.This study was arranged both in vitro and in vivo conditions.
