Obiecującą alternatywą dla obecnie stosowanych wektorów wirusowych wydają się być systemy oparte na związkach amfifilowych. Wśród nich dobrymi kandydatami są surfaktanty gemini (składające się z dwóch hydrofobowych łańcuchów i dwóch kationowych głów powiązanych przez grupę łącznikową). Badania przeprowadzono na trzech typach systemów: fosfolipid/surfaktant, fosfolipid/surfaktant/kwas nukleinowy oraz surfaktant/kwas nukleinowy (DNA i siRNA). Zostały one charakteryzowane za pomocą małokątowego rozpraszania promieniowania synchrotronowego (SAXS), różnicowej kalorymetrii skaningowej (DSC), spektroskopii podczerwieni z transformatą Fouriera (FTIR), spektroskopii dichroizmu kołowego (CD) i elektroforezy w żelu agarozowym. Badana grupa surfaktantów powoduje obniżenie temperatur przemian fazowych w układach z fosfolipidem oraz destabilizację formowanych przez niego struktur lamelarnych indukując przy tym powstawanie faz unilamelarnych, a także fazy regularnej. W układach z DNA, przy wartości stosunku ładunku dodatniego do ujemnego p/n > 1, obserwowano kondensację kwasu nukleinowego i utworzenie stabilnych kompleksów. Wykazano także zdolność surfaktantów do kompleksowania jednoniciowego i dwuniciowego oligomeru siRNA. Otrzymane wyniki pozwoliły wytypować układy będące najbardziej obiecującymi kandydatami do zastosowania w terapii genowej do transportu zarówno DNA jak i siRNA. Promising alternative to currently used virus-based vectors appear to be amphiphilic compounds. Among them, gemini surfactants (consisting of two cationic head groups and two hydrophobic tails linked by a spacer group) appear to be good candidates. This study comprised of characterization of three types of mixed systems: phoshpolipid/surfactant, surfactant/nucleic acid (both DNA and siRNA) and phospholipid/ surfactant/nucleic acid. These new delivery systems were thoroughly characterised by small angle X-ray scattering of synchrotron radiation (SAXS), differential scanning calorimetry (DSC), Fourier transform infrared spectroscopy (FTIR), circular dichroism spectroscopy (CD) and agarose gel electrophoresis. In systems with a phospholipid, the tested group of surfactants lowers the phase transitions temperatures, destabilizes the lamellar structures and induces the formation of other structural phases, mainly unilamellar structures but also a cubic phase. In systems with DNA with a charge ratio p/n > 1 (positive to negative electric charge ratio), the DNA is being condensed and stable complexes are formed. The ability of tested surfactants to complex single-stranded and double-stranded siRNA oligomers was also demonstrated.
