Mechanizmy fizjologiczne i farmakologiczne regulacji czynności miokardium chrząszczy Tenebrio molitor L. i Zophobas atratus Fab.

Abstract

W pracy badano mechanizmy fizjologiczne i farmakologiczne regulacji endogennej aktywności kurczliwej miokardium chrząszczy T. molitor i Z. atratus. Określono wpływ na kurczliwość miokardium jonów elektrogennych, blokerów jonotropowych kanałów dla Ca2+, K+ i Na+ oraz kardiotropowe działanie hormonów peptydowych (proktoliny, CCAP, Lem-MS, PK-2) i opisano modulację ich aktywności w obecności blokerów. Aktywność kurczliwą miokardium chrząszczy regulują wszystkie trzy jony. Badania farmakologiczne z blokerami jonotropowymi wykazały funkcjonowanie w błonie komórek miokardium kanałów Ca2+, K+ i Na+. Zablokowanie kanałów dla tych jonów wywołuje zróżnicowane efekty chrono- i inotropowe w akcji serca in vitro i w różny sposób zmienia częstość jego skurczów w fazie orto- i antydromowej in vivo. Endogenna aktywność kurczliwa miokardium obu chrząszczy jest stymulowana przez proktolinę i CCAP, a hamowana przez Lem-MS i PK-2. Hormony te wywołują zmiany chrono- i inotropowe w aktywności mechanicznej miokardium i modyfikują jego parametry hemodynamiczne, objętość wyrzutową, pojemność minutową i frakcję wyrzutu. Aktywność kardiotropowa hormonów jest w zróżnicowany sposób modulowana w obecności blokerów i wykazuje specyfikę gatunkową. Obecnie trudno określić mechanizm tych modulacji, czy są one efektem oddziaływania pomiędzy cząsteczką blokera i hormonu na poziomie receptora, czy kanałów jonowych. In this study, physiological and pharmacological mechanisms of endogenous contractile activity of the myocardium of beetles T. molitor and Z. atratus were investigated. Influence of electrogenic ions, ionotropic blockers for ion channels and cardiotropic actions of peptide hormones (proctolin, CCAP, Lem-MS, PK-2) were characterized. The modulation of their activity by ionotropic blockers was also described. The contractile activity of myocardium of beetles depends on three tested ions. Combinations with blockers indicate that channels for Ca2+, K+, Na+ are presented in the beetles myocardium. The block of channels for this ions caused chrono- and inotropic effects in the contractile activity in vitro and in different manner changed contractile frequency in vivo. Endogenous contractile activity of the myocardium is stimulated by proctolin and CCAP and inhibited by Lem-MS and PK-2. These hormones caused chrono- and inotropic changes in the mechanical activity of the myocardium and modified its hemodynamic parameters, stroke volume, cardiac output, ejection fraction. Cardiotropic activity of hormones was modulated, in diverse manner, in presence of ionotropic blockers and was species-specific. At present, it is difficult to determine whether this mechanism of modulation is an effect of interactions between blockers and hormones molecules on the receptor level or on ion channels level.
