We focussed on the phenomenon of the Apoptosis, a subtyp of the programmed cell death (PCD). In cells undergoing apoptosis the morphology is strongly different from those undergoing necrosis.The necrotic cell gets unstable, the membrane gets permeable and swells up bubbly by osmotic effects, finally it bursts and triggers an inflammation process in the tissue. Apoptotic cells regulate a membrane stable process under a constant use of energy by enzymes and proteasis. At our experiments with CD95 induced cell death we concentrated especially on the mitochondrial level.We analyzed the effect of 1 mm zinc in four-hour time lapse methods at the Hodgkin celline HDLM 2. After an apoptosis-triggering stimulus, the Delta Psi m of (Mitochondrial membrane potential) decreases. The release of Cytochrom C follows, which activates then over Caspase 9 the effector Caspase 3 and lets the consecutive Caspase cascade run.Membrane potential changes at the mitochondrium have to be seen as a very early sign of the apoptosis.This parameter was microscopically catched by using the indicative qualities of the JC1 flourescence and later compared with the values after zinc influence. For the first time it could be demonstrated with our work, that zinc in a concentration of 1mm has an inhibiting influence on the depolarisation of the mitochondrial Membrane Potential (Delta Psi m) in HDLM2-Cells. Morphologically these HDLM2-Cells don´t show the typical apoptosis after CD-95 and zinc stimulation. The morphology seems to be similar to mummified Hodgkin-Cells, that are an abortive kind of apoptosis. The mummified Hodgkin-Cells still show the condensation and margination processes but no more blebbing. The mummified cells may also have no mitochondrial depolarisation.
