It is commonly assumed that the inner three-dimensional structure of the collagen sponges influences cell behaviour. Until now, this influence could not be investigated systematically, since the pore structure of commercially available collagen sponges is very unregular. Therefore, a new freeze-drying process for manufacturing collagen sponges with a very homogeneous, defined and directed pore structure was developed and technically reallised. Using this process the pore size can be adjusted in the range of 20 µm to 40 µm by a variation of the solute concentration in the basic collagen suspension. The behaviour of human fibroblasts within the collagen sponges were investigated in vitro. The ex-pected influence of pore structure and size on cell behaviour could not be confirmed in case of fibro-blasts. However, fibroblast migration and proliferation could be proved over a period of 5 weeks by MTT-testing and histological staining.
